研究目的:

(1)深部与浅表刺激效应比较 基于奖赏环路的不同关键节点,本研究将比较高强度tTIS靶向下ALIC(深部白质通路)与dACC对奖赏学习及决策行为的即时影响。通过任务态功能磁共振(task-fMRI)对比不同靶点的神经反应模式,验证深部刺激是否能更有效调控皮层与皮层下奖赏通路,并评估其安全性与耐受性。 (2)奖赏环路调控机制解析 结合task-fMRI的全脑活动和功能连接分析,系统探讨不同靶点的tTIS刺激对于奖赏价值更新、行为调整与抑制控制等关键过程的神经机制。重点比较深部ALIC刺激与dACC浅表刺激在调控奖赏环路动态反应中的差异,并进一步分析这些差异如何映射到行为表现,为后续在强迫症中的靶点选择提供机制依据。  

Objectives of Study:

(1) Comparison of Deep versus Superficial Stimulation Effects Based on key nodes of the reward circuit, this study will compare the immediate effects of high-intensity tTIS targeted at the ventral ALIC (a deep white matter tract) versus the dACC on reward learning and decision-making behaviors. Task-based functional magnetic resonance imaging (task-fMRI) will be used to compare the neural response patterns under stimulation of different targets. This aims to verify whether deep stimulation can more effectively modulate cortical-subcortical reward pathways and to evaluate its safety and tolerability. (2) Elucidation of Reward Circuit Modulation Mechanisms Combining whole-brain activity and functional connectivity analyses from task-fMRI, this study will systematically investigate the neural mechanisms underlying key processes—such as reward value updating, behavioral adjustment, and inhibitory control—during tTIS stimulation of different targets. The focus will be on comparing the differences between deep ALIC stimulation and superficial dACC stimulation in modulating the dynamic responses of the reward circuit. Furthermore, these differences will be analyzed in relation to how they translate into behavioral performance, providing mechanistic insights for subsequent target selection in obsessive-compulsive disorder.

Description for medicine or protocol of treatment in detail:

 

纳入标准:

(1)年龄18–40岁的健康志愿者; (2)无精神疾病、神经系统疾病或重度躯体疾病; (3)无磁共振禁忌症,可顺利完成MRI扫描; (4)无头皮皮肤病变,能够耐受电刺激; (5)愿意完成三次独立刺激并签署知情同意书。

Inclusion criteria

1. Healthy volunteers aged 18-40 years; 2. Absence of psychiatric, neurological, or significant somatic illnesses; 3. No contraindications for MRI and ability to complete the scanning procedure successfully; 4. No scalp dermatological conditions that would preclude tolerance of electrical stimulation; 5. Willingness to complete three independent stimulation sessions and provide written informed consent.

排除标准:

(1)有癫痫、头部手术史或颅内金属植入; (2)携带心脏起搏器或其他植入式电子装置; (3)任何精神障碍史、酒精或药物滥用史; (4)妊娠、哺乳期。

Exclusion criteria:

1. History of epilepsy, cranial surgery, or presence of intracranial metal implants; 2. Bearing a cardiac pacemaker or other implanted electronic device; 3. Any history of psychiatric disorders, or alcohol or substance abuse; 4. Pregnancy or lactation.

Blinding:

A single-blind design was employed in this study. Participants were unaware of the specific stimulation condition in each experimental session. Due to the technical requirements of the procedure, the interventionist administering the stimulation could not be blinded to the condition. However, they were strictly prohibited from disclosing any information regarding the stimulation condition to the participants.
