研究目的：

本项目拟在不干预抑郁障碍患者常规治疗方案的前提下，以病例对照研究方式，以及多维度运用代谢组学、肠道菌群组学和 ERP 技术，共同探索抑郁症（MDD）患者的肠道菌群衍生 SCFAs 水平异常与 MDD 情绪加工障碍之间的关联性，通过随访比较分析抑郁障碍患者治疗前后的肠菌代谢物变化、脑电活动变化情况及其与临床症状之间的相关性，为寻找新的治疗靶点提供新的思路。  

Objectives of Study：

This project aims to explore the association between aberrant levels of gut microbiota-derived short-chain fatty acids (SCFAs) and emotional processing deficits in patients with major depressive disorder (MDD), without interfering with their routine treatment regimens. Utilizing a case-control study design and multidimensional approaches—including metabolomics, gut microbiome analysis, and event-related potential (ERP) techniques—we will longitudinally compare changes in gut microbial metabolites, electroencephalographic (EEG) activity, and their correlations with clinical symptoms before and after treatment. The findings are expected to provide novel insights into identifying potential therapeutic targets for depression.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.符合DSM-5中抑郁障碍的诊断标准（MDD组）；
2.通过半结构化临床访谈进行筛查，以排除患有精神疾病（HC组）；
3.中国汉族，男女不限；
18～65 周岁，右利手；
4.小学以上文化程度；
5.能接受脑电、抽血、留粪便等检查；
6.入组前1月内未服用过益生元、益生菌或抗生素；

Inclusion criteria

1.Meet the diagnostic criteria for depressive disorder as defined in the Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition (DSM-5) (MDD group).
2.Undergo screening via semi-structured clinical interviews to exclude subjects with psychiatric disorders (HC group).
3.Be of Chinese Han ethnicity, with no restrictions on gender.
4.Aged 18–65 years old and right-handed.
5.Have an educational level of primary school or above.
6.Be able to undergo examinations including electroencephalography (EEG), blood sampling, and fecal sample collection.
7.Have not taken prebiotics, probiotics, or antibiotics within one month prior to enrollment.

排除标准：

1.其他精神疾病（如双相障碍、精神分裂症、物质依赖） 、伴有控制不良的严 重躯体疾病，如冠心病、甲状腺功能异常等；
2.曾有过导致意识丧失的颅脑外伤史，或癫痫病史；
3.存在不能矫正至基本正常的视/听功能障碍；
4.存在明显的手指运动功能障碍（无法点击鼠标）；
5.处于妊娠或哺乳期妇女，或备孕（育）期人员；
6.近 6 月内接受过 ECT/MECT 治疗；
7.存在电极片过敏，晕血晕针；
8.不同意签署知情同意书；

Exclusion criteria：

1.Comorbidities of other psychiatric disorders (e.g., bipolar disorder, schizophrenia, substance dependence), or poorly controlled severe physical illnesses such as coronary heart disease and thyroid dysfunction.
2.A history of craniocerebral trauma leading to loss of consciousness, or a history of epilepsy.
3.Presence of uncorrectable visual or auditory impairment that cannot be ameliorated to a basically normal level.
4.Presence of obvious finger motor dysfunction (unable to operate a computer mouse).
5.Women who are pregnant or lactating, or individuals who are planning pregnancy or child-rearing.
6.Having received electroconvulsive therapy (ECT)/modified electroconvulsive therapy (MECT) within the past 6 months.
7.A history of allergies to electrode patches, or a phobia of blood or needle sticks.
8.Refusal to sign the informed consent form.
