研究目的:

1.本研究拟构建臭氧暴露染毒舱系统,采用随机对照设计,揭示臭氧急性暴露的潜在健康风险,为制定空气质量标准与人群防护策略提供依据; 2.识别臭氧暴露对于cfDNA片段大小,甲基化水平等特征的影响,并探索其特征变化的毒理学意义,评估其表征臭氧健康损害效应的可行性。  

Objectives of Study:

1. This study aims to develop an ozone exposure chamber system and employs a randomized controlled design to elucidate potential health risks from acute ozone exposure, providing evidence for establishing air quality standards and population protection strategies. 2. The research will identify ozone-induced alterations in cfDNA characteristics including fragment size and methylation levels, investigate the toxicological significance of these changes, and evaluate the feasibility of utilizing cfDNA features to characterize ozone-related health damage.

Description for medicine or protocol of treatment in detail:

 

纳入标准:

(1)18~26岁; (2)汉族; (3)实验期间均住在广州; (4)能够完成整个实验; (5)BMI位于18.5-24.9之间。

Inclusion criteria

(1) Aged 18–26 years; (2) Han Chinese ethnicity; (3) Resided in Guangzhou during the experimental period; (4) Able to complete the entire experiment; (5) Body Mass Index (BMI) between 18.5 and 24.9.

排除标准:

(1)吸烟或酗酒史; (2)8周内服用过药物或膳食补充剂; (3)患有过敏性疾病的受试者,如过敏性鼻炎、过敏性哮喘; (4)患有心血管疾病的受试者,如先天性心脏病、肺心脏病和高血压; (5)患有呼吸道疾病的受试者,如哮喘、慢性支气管炎和慢性阻塞性肺病; (6)患有慢性疾病的受试者,如糖尿病、慢性肝炎和肾脏疾病; (7)有大手术史的受试者; (8)有精神疾病的受试者,如抑郁,癫痫等; (9) 肺活量检测异常的受试者(FEV1和FVC≤预测的75%,FEV1/FVC ≤ 0.65)。

Exclusion criteria:

(1) History of smoking or alcohol abuse; (2) Used medications or dietary supplements within 8 weeks; (3) Subjects with allergic diseases, such as allergic rhinitis and allergic asthma; (4) Subjects with cardiovascular diseases, such as congenital heart disease, pulmonary heart disease, and hypertension; (5) Subjects with respiratory diseases, such as asthma, chronic bronchitis, and chronic obstructive pulmonary disease (COPD); (6) Subjects with chronic diseases, such as diabetes mellitus, chronic hepatitis, and kidney disease; (7) Subjects with a history of major surgery; (8) Subjects with psychiatric disorders, such as depression, epilepsy, etc.; (9) Subjects with abnormal pulmonary function test results (Forced Expiratory Volume in 1 second [FEV1] and Forced Vital Capacity [FVC] <= 75% of the predicted values, and FEV1/FVC <= 0.65).

由独立人员通过R Studio编写随机化程序。从CSV文件加载30名受试者ID信息,为确保结果的可复现性,使用set.seed(123)设定随机种子。
