构建国人成人不同类型的白内障专病数据库,并以此为基础探索不同类型成人白内障临床特征、分子遗传学差异、代谢组学差异,建立及完善不同类型白内障的生物样本库。  

Objectives of Study:

To construct a specialized disease database for different types of cataracts in Chinese adults. Building on this foundation, we will explore clinical characteristics, molecular genetic variations, and metabolomic differences among various types of adult cataracts, and establish and refine a specialized biobank for different cataract types.

纳入标准:

1.年龄在18周岁或以上;
2.单眼或双眼晶状体混浊,并且拟行白内障手术;
3.同意参与本研究,且签署知情同意书;

Inclusion criteria

1. Aged 18 years or older; 2. Presence of unilateral or bilateral lens opacity scheduled for cataract surgery; 3. Willingness to participate in the study with signed informed consent form;

排除标准:

1.不拟行白内障手术;

数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC:

数据收集 1)眼部状况:眼前段照相等 2)生物样本采集:收集受试者术前血液样本、术中剥离的前囊膜组织、抽取前房水、晶体皮质、核等,由专人将样品冻存于区庄院区三号楼五楼公共平台实验室,以便进一步实验分析 3)基因测序:基因测序将在与无外资合作的第三方检测公司签订检测合同后,交由第三方检测公司进行。受试者的术前血液样本将进行全外显子测序,捕获受试者全基因组外显子区域DNA序列并进行高通量测序,受试者将获得一份检测报告(数据文件大小约2M/样本)。术中剥离的前囊膜组织、晶状体皮质、核将进行RNA测序,研究者获得原始测序文库(数据文件大小约6G/样本)。 4)代谢组学检测:代谢组学检测将在与无外资合作的第三方检测公司签订检测合同后,由第三方检测公司进行。受试者术中抽取前房水将通过超高效液相色谱-高分辨质谱联用技术进行代谢组分析,研究者获得原始定性定量代谢谱(数据文件大小约1G/样本)。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture:

Data Collection 1) Eye condition: Front segment photography, etc 2) Biological sample collection: Collect preoperative blood samples from the subjects, dissect the anterior capsule tissue during surgery, extract anterior chamber water, crystalline cortex, nucleus, etc., and have a dedicated person freeze the samples in the public platform laboratory on the fifth floor of Building 3 in the district for further experimental analysis 3) Gene sequencing: Gene sequencing will be conducted by a third-party testing company after signing a testing contract with a non foreign-owned testing company. The preoperative blood sample of the subject will undergo whole exome sequencing to capture the DNA sequence of the subject's whole genome exon region and perform high-throughput sequencing. The subject will receive a test report (data file size of approximately 2M/sample). The anterior capsule tissue, lens cortex, and nucleus removed during surgery will undergo RNA sequencing, and the researchers will obtain the original sequencing library (data file size of approximately 6G/sample). 4) Metabolomics testing: Metabolomics testing will be conducted by a third-party testing company without foreign investment cooperation after signing a testing contract. The extraction of anterior chamber water from the subjects during surgery will be analyzed for metabolomics using ultra-high performance liquid chromatography high-resolution mass spectrometry. The researchers will obtain the original qualitative and quantitative metabolic profiles (data file size of approximately 1GB/sample).
