研究目的: |
1)采集耐药性颞叶癫痫-海马硬化患者切除的海马标本,通过病理、激光捕获显微解剖和质谱技术的结合,分离出海马CA1、CA2、CA3、CA4、DG、SUB六个区域的蛋白,绘制出耐药性颞叶癫痫-海马硬化患者海马的空间蛋白质图谱。阐明海马硬化不同区域的蛋白质表达特点及各解剖区域的主要功能。2)利用生物信息学,进一步寻找与神经细胞(神经元、星形胶质细胞、小胶质细胞)变化相关的蛋白质谱及生物过程。筛选介导海马硬化的分子事件,为其潜在的药物靶点提供有用的信息,控制耐药性癫痫患者的癫痫发作。 |

Objectives of Study: |
1)Collect the hippocampal specimens resected from patients with drug-resistant temporal lobe epilepsy-hippocampal sclerosis. Through a combination of pathology, laser-capture microdissection, and mass spectrometry, protein of six regions of hippocampal, including CA1, CA2, CA3, CA4, DG, SUB, are isolated and produced a spatial region-resolved proteome map of the hippocampal of patients with drug-resistant temporal lobe epilepsy-hippocampal sclerosis. Which will clarify the proteomic characteristics of six regions of hippocampal and the main functions of each region in hippocampal sclerosis. 2)Then find the protein profile and biological processes related to the changes of the nerve cells (neurons, astrocytes, microglia)?using the bioinformatics?method. The proteins were used to explore the molecular events mediating hippocampal sclerosis, which would provide useful information for the potential drug target and control seizures of patients with drug-resistant epilepsy. |

纳入标准: |
1)符合2010 年国际抗癫痫联盟(International League Against Epilepsy,ILAE)定义的耐药性癫痫,年龄14-60岁,男女不限。 2)通过神经影像学、脑电监测或功能成像,确定了致痫区,为内侧颞叶癫痫。 3)术后病理结果提示海马硬化。 4)获得医院伦理委员会批准,患者知情并同意。 |

Inclusion criteria |
1)Meet the definition criteria defined by the 2010 International League Against Epilepsy (ILAE): Drug resistant epilepsy. 2)The epileptogenic zone was identified as temporal lobe epilepsy through neuroimaging, EEG monitoring, or functional imaging. 3) Postoperative pathological analysis reveals hippocampal sclerosis. 4)Obtain the approval from the hospital ethics committee, and with the informed consent of patients before inclusion. |

排除标准: |
1)通过神经影像学、脑电监测或功能成像,致痫区不属于内侧颞叶癫痫的。 2)术后病理结果提示肿瘤、皮质发育不良等其他病因所致的耐药性癫痫。 3)脑组织的海马结构不完整的样本。 |

Exclusion criteria: |
1)The epileptogenic zone does not meet the criteria of temporal lobe epilepsy through neuroimaging, EEG monitoring, or functional imaging. 2)Postoperative pathological analysis reveals drug-resistant epilepsy caused by other etiologies such as tumors and cortical dysplasia. 3) The hippocampal formation of the brain sample is incomplete. |
