研究目的: |
本研究通过详细采集KCNQ2变异儿童的临床表型,通过PCR-NGS区分嵌合体与杂合体,并构建病人来源突变相关质粒,进行HEK293细胞转染后采集全细胞膜片钳单通道电生理特征数据,并采集入组患儿的高密度头皮脑电图、脑磁数据及核磁数据,对脑电、脑磁原始数据进行全脑可视化源定位处理,并对不同脑电、脑磁通道进行预处理和数据提取,通过时间序列分析、谱分析等方式对脑电、磁改变及其不同区域的波谱常见神经递质半定量分析,对不同脑电振荡模式、神经递质与突变通道的电特性进行关联分析,通过对详细临床表型、在体脑电磁特征及突变电生理特征的完整描述及它们之间的相关关系,获得M电流异常对不同脑电振荡及脑区的影响和它们与认知的关系,探索新型神经生物电生理标志物,为进一步的机制研究与神经调控治疗研究提供依据。 |

Objectives of Study: |
In this study, the clinical phenotype of children with KCNQ2 variants was collected in detail, the chimeras and heterozygotes are distinguished using PCR-NGS, the electrophysiological characteristics of the channel were collected after the transfection of the HEK293 engineered cell line by constructing mutation-related plasmids, and the high-density scalp EEG data, MR and MEG of the enrolled children were collected, the original EEG and MEG data were preprocessed and extracted, and the EEG/MEG changes and their effects on different brain oscillation patterns were analyzed by time series analysis and spectral analysis, and the correlation analysis was carried out in combination with the electrophysiological changes of specific variants. Through the complete description of detailed clinical phenotypes, EEG/MEG characteristics and mutant electrophysiological characteristics and the correlation between them, the effects of M current abnormalities on different EEG oscillations and brain regions and their relationship with cognition were obtained, which provided a basis for further mechanism research |
