SLE发病机制仍十分不清楚,通过本项目可以: 1. 获取SLE发生、发展(脏器受累)以及不同治疗反应等多阶段的单细胞转录组、单细胞空间转录组、免疫组库测序数据,建立多维度、标准化的SLE免疫力大数据及特征图谱,推动SLE早期诊断及个体化精准治疗。 2. 从分子、细胞和组织层面数字化解码SLE全疾病周期的核心免疫力特征,结合患者的临床特征对数据进行重编码,最终建立SLE的免疫力数字化解码/重编码模型,并完成可视化软件呈现(如SLE-Talk)。  

Objectives of Study:

The pathogenesis of systemic lupus erythematosus (SLE) remains poorly understood. Through this project, we aim to: Establish a Multi-Dimensional SLE Immune Atlas Generate single-cell transcriptomic, spatially resolved transcriptomic, and immune repertoire sequencing data across key stages of SLE progression—including disease onset, organ involvement, and differential treatment responses. This will enable the construction of a standardized, multi-dimensional immune profiling database to advance early diagnosis and personalized precision therapy for SLE. Decode and Re-Encode SLE Immunity via Digital Modeling Systematically decode core immune signatures of SLE at molecular, cellular, and tissue levels throughout the disease continuum. By integrating these multi-omics data with clinical profiles, we will develop a digital immune decoding/re-encoding model for SLE and implement it as an interactive visualization platform (e.g., SLE-Talk), facilitating translational applications.

纳入标准:

1.纳入的SLE患者需符合1997或2019 EULAR系统性红斑狼疮分类诊断标准。同时还纳入SLE遗传易感人群(一代直系亲属患有SLE,本人尚无任何症状,自身抗体均阴性),SLE临床前期人群(有临床症状或体征,但不满足SLE分类标准的患者),以及符合1997年类风湿关节炎分类标准的RA患者,符合2012年SLICC干燥综合征分类标准的pSS患者。

Inclusion criteria

1.SLE: Must meet either the 1997 ACR or 2019 EULAR/ACR classification criteria for systemic lupus erythematosus. High-risk lupus: First-degree relatives of SLE patients who are currently asymptomatic and test negative for all autoantibodies. Early lupus: Subjects with clinical symptoms/signs suggestive of SLE but not fulfilling the classification criteria. Disease controls: Rheumatoid arthritis (RA) patients meeting the 1997 ACR classification criteria for RA. Primary Sj?gren’s syndrome (pSS) patients fulfilling the 2012 SLICC classification criteria for pSS.

1.排除年龄小于18岁,且合并其他危重疾病的患者(如感染、心脏疾病、肺脏疾病、肝脏疾病等)。

Exclusion criteria:

1.Age <18 years. Comorbid severe systemic diseases (e.g., active infections, cardiovascular, pulmonary, hepatic, or other major organ dysfunction).

数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC:

SLE专病随访门诊中完成患者的临床和生活质量评估、治疗方案调整、生物标本留取、随访患者的管理和病历资料保存工作。多组学测序数据保存在我科专用服务器中,并在研究结束时上传至中国国家生物信息中心存储。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture:

The clinical and quality-of-life assessment of patients, adjustment of treatment plans, collection of biological samples, management of follow-up patients, and preservation of medical records are completed in the SLE-specialized follow-up clinic. Multi-omics sequencing data are stored in the dedicated server of our department, and will be uploaded to the National Genomics Data Center of China for storage at the end of the study.
