研究目的:

1.阐明关联机制:探究肌肉流失、骨密度/骨质量改变与脑认知衰退之间的分子及病理生理联系; 2.发现预警标志物:基于患者血液/术中标本/影像数据筛选跨系统早期预测指标; 3.支撑精准干预:为开发靶向肌-骨-脑共病的防治策略提供循证依据和资源平台。通过标准化采集临床数据、影像资料和生物样本,助力转化医学研究。  

Objectives of Study:

1. Elucidating the association mechanism: exploring the molecular and pathophysiological links between muscle loss, bone density/bone mass changes and brain cognitive decline. 2. Early warning markers: screening early predictive indicators across systems based on patient blood/intraoperative specimens/image data; 3. Support precision intervention: provide evidence-based basis and resource platform for the development of prevention and treatment strategies targeting muscle-bone-brain comorbidities. Transform translational medicine research through standardized collection of clinical data, imaging data, and biological samples.

纳入标准:

1.疾病诊断明确:符合国际或国内诊断标准的以下疾病之一:肌少症(AWGS 2019标准);骨质疏松症(DXA骨密度T值<=-2.5或脆性骨折史);强直性脊柱炎(ASAS分类标准);黄韧带骨化(OLF)或椎间盘突出(MRI/CT确诊);阿尔茨海默病(AD)(NIA-AA诊断标准); 2.年龄范围:大于等于18岁; 3.知情同意:自愿签署知情同意书,配合随访及样本采集。

Inclusion criteria

1. Clear disease diagnosis: one of the following diseases that meet international or domestic diagnostic criteria: sarcopenia (AWGS 2019 standard); Osteoporosis (DXA bone density T-value <=-2.5 or history of fragility fracture); Ankylosing spondylitis (ASAS classification criteria); ossification of the ligamentum flavum (OLF) or herniated disc (confirmed by MRI/CT); Alzheimer's disease (AD) (NIA-AA diagnostic criteria); 2. Age range: greater than or equal to 18 years old; 3. Informed consent: voluntarily sign the informed consent form and cooperate with follow-up and sample collection.

排除标准:

1.严重合并症:恶性肿瘤(5年内活动性肿瘤,非黑色素瘤皮肤癌除外);终末期器官衰竭(如心功能NYHA IV级、eGFR <30 mL/min);未控制的感染或免疫缺陷疾病(如HIV/AIDS); 2.有药物、酒精或毒品依赖者; 3.有严重精神类疾病者。

Exclusion criteria:

1. Severe comorbidities: Malignant tumors (active tumors within 5 years, excluding non-melanoma skin cancer); End-stage organ failure (such as NYHA class IV cardiac function, eGFR < 30 mL/min); Uncontrolled infections or immunodeficiency diseases (such as HIV/AIDS); 2. Those with drug, alcohol or substance dependence; 3. Those with severe mental disorders.

数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC:

1. 数据管理 使用微软Excel软件对收集的患者数据进行初步整理和存储。 2. 统计分析 将数据导入Graphpad Prism 9进行分析。数据以平均数±标准差、频率和百分比表示,视情况而定。变量间的相关性采用Spearman秩相关系数进行分析。通过构建受试者工作特征(ROC)曲线来评估试验之间的相关性。p值< 0.05认为有统计学意义。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture:

1. Data Management The collected patient data were initially organized and stored using Microsoft Excel software. 2. Statistical Analysis The data were imported into Graphpad Prism 9 for analysis. The data were presented as mean ± standard deviation, frequency, and percentage, depending on the circumstances. The correlation between variables was analyzed using the Spearman rank correlation coefficient. The correlation between the tests was evaluated by constructing receiver operating characteristic (ROC) curves. A p value < 0.05 was considered statistically significant.
