研究目的：

本研究旨在评估激光预处理联合ALA-PDT治疗口腔黏膜白斑的有效性和安全性，对比其与传统PDT治疗在疗效、治疗次数、单位病损面积治疗时长、不良反应等方面的差异，为临床治疗提供新选择。同时通过动物模型进一步验证，通过激光预处理，能够使得ALA更容易渗透组织。  

Objectives of Study：

This study aims to evaluate the efficacy and safety of laser pretreatment combined with ALA-PDT in the treatment of oral mucosal leukoplakia, compare the differences between it and traditional PDT treatment in terms of efficacy, treatment frequency, treatment duration per unit lesion area, adverse reactions, etc., and provide a new option for clinical treatment. At the same time, it was further verified through animal models that laser pretreatment can make ALA more easily penetrate tissues.

纳入标准：

① 年龄 18-75 岁，性别不限，中国汉族。 ②符合口腔白斑病 (OLK)诊断标准： ③ 发生于口腔黏膜上以白色为主的损害,不能擦去 ,也不能以临床和组织 病理学的方法诊断为其他可定义的损害 ,属于癌前病变或潜在恶性疾患范 畴 ,不包括吸烟、局部摩擦等因素去除后可以消退的口腔白角化症。 ④ 为保证治疗一致性，病损最大径不超过 4cm。仅纳入均质白斑，疣状白 斑及颗粒白斑。

Inclusion criteria

1 Age: 18-75 years old, gender not limited, Han Chinese. 2 Meet the diagnostic criteria for oral leukoplakia (OLK) : 3 Lesions that occur on the oral mucosa and are mainly white cannot be wiped off, nor can they be clinically or histologically observed Pathological methods diagnose it as other definable lesions, which fall within the category of precancerous lesions or potential malignant disorders This does not include oral leukokeratosis that can subside after the removal of factors such as smoking and local friction. 4 To ensure the consistency of treatment, the maximum diameter of the lesion should not exceed 4cm. Only homogeneous leukoplakia and verrucous leukoplakia were included Spots and granular white patches.

排除标准：

① 妊娠期、哺乳期妇女、1 年内有妊娠计划的育龄妇女。 ②伴有其它口腔黏膜疾患 ③ 伴全身严重急性感染性疾病;未有效控制的系统性疾病 (凝血功能障碍、 自身免疫性疾病、糖尿病、心血管疾病、精神病等 )；患有其他部位恶性 肿瘤者。 ④ 既往因口腔潜在恶性疾病或头颈部肿瘤接受过外科治疗或放化疗的 患者。 ⑤ 对 630±5nm 左右波长范围光过敏者。 ⑥ 对局部用盐酸氨酮戊酸溶液中任何成分过敏者。 ⑦ 局麻药物过敏者。 ⑧ 卟啉过敏者。 ⑨ 任何其他原因不愿或无法接受本项治疗的患者。

Exclusion criteria：

1 Pregnant women, lactating women, and women of childbearing age with pregnancy plans within one year. 2 Accompanied by other oral mucosal disorders 3 Accompanied by severe acute infectious diseases throughout the body; Uncontrolled systemic diseases (coagulation disorders, autoimmune diseases, diabetes, cardiovascular diseases, mental illnesses, etc. Suffering from malignancy in other parts of the body. 4 Those who have received surgical treatment or radiotherapy and chemotherapy for potential malignant oral diseases or head and neck tumors in the past patient. 5 People who are allergic to light within a wavelength range of approximately 630±5nm. 6 Those who are allergic to any component in the topical solution of aminovaleric hydrochloride. 7 People who are allergic to local anesthetic drugs. 8 People allergic to porphyrins. 9 Any patient who is unwilling or unable to receive this treatment for any other reason.

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：

每位就诊患者均建电子病历在我院存档，便于随时调取。 研究相关数据采取纸质版及六元空间电子版同时录入的方法。研究者根据受试者的原始观察记录，将数据及时、完整、正确、清晰地载入原始数据。录入采用相应的数据库系统双人双机录入。 原始数据在按要求完成数据录入和核查后，按编号的顺序归档保存，并填有检索目录等，以备查考。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：

Each patient seeking medical treatment will have an electronic medical record archived in our hospital for easy access at any time. The relevant research data was entered simultaneously in both paper and electronic versions of Liuyuan Space. Based on the original observation records of the subjects, the researchers promptly, completely, accurately and clearly loaded the data into the original data. Input is carried out using the corresponding database system with two people and two machines. After the original data is entered and verified as required, it is archived and saved in the order of their numbers, and a search directory and other information are filled in for future reference.
