结直肠癌(CRC)中微卫星稳定型(MSS)占85%,其CD8+T细胞浸润和活性低,被视为“冷肿瘤”,对PD-1抗体等免疫疗法响应不佳,预后较差。作为肿瘤微环境的重要组分,肠道菌群失调被认为是MSS型CRC对免疫治疗耐受的关键因素。本研究从“微生物-代谢-免疫”综合视角,期望为开发以靶向肠道微生物-提升肿瘤“热度”-增强CRC免疫疗效的策略提供理论和临床应用新思路。 |

Objectives of Study: |
Microsatellite-stable (MSS) colorectal cancer (CRC) accounts for approximately 85% of all CRC cases. These tumors are characterized by low CD8? T cell infiltration and activity, rendering them “cold tumors” that respond poorly to immune checkpoint blockade therapies such as anti-PD-1 antibodies, and are associated with unfavorable clinical outcomes. As a critical component of the tumor microenvironment, gut microbiota dysbiosis is increasingly recognized as a key factor contributing to the immunotherapy resistance observed in MSS CRC. This study adopts an integrated "microbiota–metabolite–immunity" perspective, aiming to provide new theoretical insights and translational strategies for converting cold tumors into hot ones by targeting the gut microbiota to enhance the immune responsiveness of CRC. |

1.微卫星稳定型(MSS)结直肠癌(CRC)患者; 1) 经病理学确诊为 MSS 型 CRC; 2) 年龄在 18-75 岁之间; 3) 未接受任何抗肿瘤治疗(包括手术、 化疗、 放疗、 免疫治疗等)。 2.健康受试者; 1) 年龄、 性别与患者组匹配; 2) 无 CRC 病史或家族史; 3) 无其他可能明显影响肠道健康的重大疾病史(如炎症性肠病) 3.自愿参加研究并签署知情同意书; 4.能够积极配合完成样本采集及问卷调查; |

Inclusion criteria |
1.Patients with microsatellite-stable (MSS) colorectal cancer (CRC); 1) Pathologically confirmed diagnosis of MSS type CRC; 2) Be between the ages of 18-75 years; 3) Not receiving any anti-tumor therapy (including surgery, chemotherapy, radiotherapy, immunotherapy, etc.). 2.Healthy volunteers; 1) Age, gender matched to patient group; 2) no history or family history of CRC; 3) No history of other major diseases that may significantly affect gut health (e.g., inflammatory bowel disease) 3.Willing to participate in the study and sign the informed consent form; 4.Able to actively cooperate with sample collection and questionnaire completion; |
