研究目的：

1） 选择亚甲基四氢叶酸还原酶（MTHFR）（C677T）rs1801133、MTHFR （A1298C）rs1801131、乙醛脱氢酶2（ALDH2） rs 671、载脂蛋白E（APOE） rs 429358、APOE526 rs 7412 及水通道蛋白4（AQP4）rs9951307脑卒中相关基因位点，分析其多态性在岛城PS高危人群中的频数分布； 2） 建立岛城麻醉手术患者临床资料联合关键基因多态性数据库； 3） 基于患者临床资料和关键基因多态性数据，构建PS多参数预测模型体系，建立PS防治体系。  

Objectives of Study：

1. Select stroke-related gene loci including methylenetetrahydrofolate reductase (MTHFR) (C677T) rs1801133, MTHFR (A1298C) rs1801131, aldehyde dehydrogenase 2 (ALDH2) rs671, apolipoprotein E (APOE) rs429358, APOE rs526 rs7412, and aquaporin 4 (AQP4) rs9951307, and analyze the frequency distribution of their polymorphisms in the high-risk population for perioperative stroke (PS) in Daocheng Island City. 2. Establish a database combining clinical data with key gene polymorphism data for anesthesia and surgical patients in Daocheng Island City. 3. Based on patients' clinical data and key gene polymorphism data, construct a multi-parameter prediction model system for PS and establish a PS prevention and treatment system.

纳入标准：

北方汉族；根据基于临床数据的PS预测模型，Stroke after surgery tool或Mobile web-based, cardiac, stroke mortality risk model，预测PS发生率≥1%的患者；预计麻醉手术时长≥2小时的择期手术患者；

Inclusion criteria

Northern Han Chinese; identify patients with a predicted incidence of perioperative stroke (PS) >=1% according to clinical data-based PS prediction models, such as the Stroke after Surgery tool or the Mobile web-based, cardiac, stroke mortality risk model; and select patients undergoing elective surgery with an expected anesthesia and surgery duration of >=2 hours.

排除标准：

口腔黏膜溃疡；口腔、咽部、舌肿瘤等；有出血倾向，采集口腔黏膜标本可能出血者；不能配合采集口腔黏膜脱落细胞者（如合并精神疾患或认知功能障碍）；近一个月内参加过临床实验；未成年人、孕产妇及哺乳期妇女；伴有其他情况，研究者认为不适合入选的患者。

Exclusion criteria：

Oral mucosal ulcer; tumors of the oral cavity, pharynx, and tongue, etc.; individuals with a tendency to bleed or for whom collecting oral mucosal specimens may result in bleeding; those unable to cooperate with the collection of oral mucosal exfoliated cells (such as those with concurrent mental illness or cognitive impairment); individuals who have participated in clinical trials within the past month; minors, pregnant and lactating women; patients with other conditions deemed unsuitable for inclusion by the investigator.
