首先通过临床特征确定ALS研究病例,然后通过对ALS病人以及家属进行全基因组测序,确定导致疾病的已/未报道突变类型。之后通过直接神经谱系重编程方法,构建ALS患者来源诱导神经干细胞稳定细胞系,开发更真实细胞以及脑类器官ALS疾病模型,以期反映出已知以及探索未知ALS细胞病理特征,并应用于药物筛选与研发。 |

Objectives of Study: |
First, ALS research cases are identified through clinical manifestations. Then, through whole-genome sequencing of ALS patients and their families, the types of mutations that cause or do not cause the disease are determined. After that, ALS patient-derived induced pluripotent stem cell (iPSC) stable cell lines are established using direct neurogenic lineage reprogramming methods, and more realistic cell and brain-like organ models are developed to reflect known and explore unknown ALS cellular pathological features, which can be used for drug screening and development. |

1.符合 2008 年 Awaji-Shima 诊断标准中的临床确诊 ALS 患者; 2.患者及其家属理解并同意进行临床资料和外周血样本采集。 |
