研究目的：

收集350例志愿者的尿液样本进行甲基化检测，结合临床资料，构建基于尿液甲基化的膀胱癌无创诊断模型。 进一步探究尿液脱落细胞目标基因甲基化特征是否与膀胱癌肌层浸润相关。  

Objectives of Study：

Urine samples of 250 volunteers were collected for methylation detection. Combined with clinical data, a non-invasive diagnosis model of bladder cancer based on urine methylation was established. To further explore whether the methylation characteristics of target genes in urine exfoliated cells are related to muscular infiltration of bladder cancer.

纳入标准：

健康志愿者组： 1、年龄>18 岁，男女不限。 2、 同意签署《科研知情同意书》。 3、未患泌尿系统疾病。 患者组： 1、年龄>18 岁，男女不限。 2、自研究开始，来我科就诊，并被初诊为膀胱癌、肾癌、前列腺癌、输尿管癌、肾盂癌、输尿管炎、尿路上皮增生、膀胱无病变的患者。 3、同意签署《科研知情同意书》

Inclusion criteria

Healthy Volunteer Group: 1. Age> 18 years old, male or female. 2. Agree to sign the "Informed Consent Form for Scientific Research". 3. No urinary system diseases. Patient group: 1. Age> 18 years old, male or female. 2. Since the beginning of the study, patients who have come to our department for treatment and have been newly diagnosed as bladder cancer, kidney cancer, prostate cancer, ureteral cancer, renal pelvic cancer, ureteritis, urothelial hyperplasia, and bladder lesions. 3. Agree to sign the "Informed Consent Form for Scientific Research"

排除标准：

1、拒绝签署《科研知情同意书》。 2、近期已行化疗、免疫治疗、手术治疗者。 3、无法获得足够尿液者。

Exclusion criteria：

1. Refuse to sign the "Informed Consent for Scientific Research". 2. Those who have recently undergone chemotherapy, immunotherapy and surgery. 3. Those who are unable to obtain enough urine.

测量方法：

将提取的基因组DNA进行亚硫酸氢盐转化，未发生甲基化的胞嘧啶被转化为尿嘧啶。而甲基化的胞嘧啶不变。然后将亚硫酸氢盐转化的DNA(Bis-DNA)做PCR扩增，检测目的基因的片段是否扩增。

The extracted genomic DNA was Bisulfite transformed and the unmethylated cytosine was converted to uracil. The methylated cytosine is unchanged. The Bisulfite transformed DNA (bis-DNA) was then amplified by polymerase chain reaction (PCR) to determine whether the target gene fragment was amplified.
