研究目的:

本研究的主要目的是探究不同年龄和性别人类脑脊液中蛋白质和代谢物的组成差异,验证中枢神经系统(脑和脊髓)衰老的新标志物(TREM2和CHIT1)在不同年龄、性别人脑脊液中的表达趋势,揭示人中枢神经系统衰老的特征,挖掘新的脑脊液标志物并作相关的机制验证。基于脑脊液的代谢物和蛋白质表达特征,进一步筛选脑脊液生物标志物并构建中枢神经系统衰老评分模型,为中枢神经系统生物学年龄评估、病理性中枢神经系统衰老的早期诊断、治疗以及疗效评价提供参考依据。  

Objectives of Study:

The main purpose of this study was to explore the differences in the composition of proteins and metabolites in human cerebrospinal fluid of different ages and genders, verify the expression trend of new markers of aging of the central nervous system (brain and spinal cord) (TREM2 and CHIT1) in the cerebrospinal fluid of people of different ages and genders, and reveal the characteristics of aging of the human central nervous system. To explore new cerebrospinal fluid markers and validate the related mechanisms. Based on the expression characteristics of metabolites and proteins in cerebrospinal fluid, further screening of cerebrospinal fluid biomarkers and construction of central nervous system aging score model provide reference for biological age assessment of central nervous system, early diagnosis, treatment and efficacy evaluation of pathological central nervous system aging.

Description for medicine or protocol of treatment in detail:

 

纳入标准:

1)受试者的年龄在相应区间内:青年组(18-35岁)、壮年组(36-45岁)、中年组(46-55岁)、中老年组(56-65岁)、老年组(66-80岁)和高龄组(≥81岁); 2)受试者无中枢神经系统器质性疾病或中枢神经症状,如:头晕、嗅觉丧失、味觉丧失、认知障碍等; 3)受试者无神经精神功能障碍综合征,如:注意力、集中力、记忆力、信息处理速度和执行功能受损,或抑郁、焦虑、睡眠障碍等神经精神症状; 4)受试者的临床资料或检验学资料齐全; 5)受试者本人或监护人/法定代理人自愿配合该研究并签署知情同意书。

Inclusion criteria

1) The age of the subjects was within the corresponding range: young group (18-35 years old), mature group (36-45 years old), middle-aged group (46-55 years old), middle-and-old-aged group (56-65 years old), elderly group (66-80 years old) and senior group (≥81 years old); 2) The subjects have no organic diseases of the central nervous system or central nervous symptoms, such as dizziness, loss of smell, loss of taste, cognitive impairment, etc.; 3) The subjects have no neuropsychiatric syndrome, such as impaired attention, concentration, memory, information processing speed and executive function, or neuropsychiatric symptoms such as depression, anxiety and sleep disorders; 4) The clinical data or laboratory data of the subject are complete; 5) The subject himself or his guardian/legal representative voluntarily cooperate with the study and sign the informed consent.

排除标准:

1)受试者入院后检查提示严重系统性疾病,如:高血压(2-3级)、糖尿病、心脑血管病、风湿免疫病等,或处于急、慢性感染性疾病的活动期; 2)受试者患有严重中枢神经系统器质性疾病,如:颅内肿瘤、脑出血、蛛网膜下腔出血等; 3)受试者患有或曾患有精神性疾病,如:抑郁症、精神分裂症等; 4)受试者既往有颅脑手术史; 5)受试者既往有恶性肿瘤史; 6)受试者处于怀孕期或哺乳期。

Exclusion criteria:

1) After admission, the examination of the subjects indicated serious systemic diseases, such as hypertension (grade 2-3), diabetes, cardiovascular and cerebrovascular diseases, rheumatological and immunological diseases, etc., or were in the active stage of acute or chronic infectious diseases; 2) The subjects had severe organic diseases of the central nervous system, such as intracranial tumors, cerebral hemorrhage, subarachnoid hemorrhage, etc.; 3) The subject is or has been suffering from mental illness, such as depression, schizophrenia, etc.; 4) The subjects have a history of craniocerebral surgery; 5) The subject has a history of malignant tumors; 6) The subject is pregnant or breastfeeding.
