研究目的：

1. 通过单细胞RNA测序技术，绘制人类妊娠早期母胎界面细胞图谱，寻找RSA患者与正常妊娠女性母胎界面免疫细胞相关的差异转录基因，借助空间转录组平台，对母胎界面细胞图谱进行验证，观测子宫蜕膜中不同部位功能区基因表达的差异，构建母胎免疫细胞通讯网络，并寻找与蜕膜巨噬细胞相关的关键转录基因及信号转导通路。 2. 探索RSA患者蜕膜巨噬细胞增殖、凋亡水平、细胞吞噬功能、胞葬功能及M1、M2型巨噬细胞极化路径相关因子表达差异 通过观察RSA患者与正常妊娠女性蜕膜组织辅助性T细胞Th1、Th2数量，巨噬细胞M1、M2型细胞数量及巨噬细胞M1、M2极化路径相关因子PERK、ATF-4、PSAT1、α-KG、p-JAK、p-STAT1表达水平、蜕膜巨噬细胞增殖、凋亡、细胞吞噬、胞葬功能，探索RSA患者蜕膜巨噬细胞功能及极化路径相关因子表达水平的差异。  

Objectives of Study：

1. Single cell RNA sequencing technology was used to map the maternal/fetal interface cell map in the early stage of human pregnancy, search for differential transcriptional genes related to maternal/fetal interface immune cells in RSA patients and normal pregnant women, verify the maternal/fetal interface cell map with the help of spatial transcriptome platform, and observe the differences in gene expression in functional regions of different parts of decidua. The communication network of maternal-fetal immune cells was constructed, and the key transcription genes and signal transduction pathways related to decidual macrophages were searched. 2. To explore the differences in the proliferation and apoptosis levels of decidual macrophages in RSA patients, cytophagocytosis and cytoburial functions, and the expression of factors related to the polarization pathway of M1 and M2 macrophages. By observing the number of helper T cells Th1 and Th2 in decidual tissue between RSA patients and normal pregnant women, The number of macrophage M1 and M2 type cells, the expression levels of macrophage M1, M2 polarization path-related factors PERK, ATF-4, PSAT1, α-KG, p-JAK, p-STAT1, the proliferation, apoptosis, phagocytosis, and cytoburial function of decidual macrophages, To explore the differences in the function of decidual macrophages and the expression levels of polarization pathway-related factors in RSA patients.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.复发性流产纳入标准：与同一配偶连续发生2次及以上在妊娠28周之前的妊娠丢失，包括生化妊娠；临床症状有停经史，少量或无阴道流血，B超提示宫内妊娠，无胚芽或无心管搏动。
2.对照组纳入标准：孕12w内的非医学原因行人工流产的正常妊娠女性；与早期RSA患者胎龄相匹配；至少于足月时分娩一名健康婴儿；B超提示胚胎发育无异常，有胚芽和心管搏动。

Inclusion criteria

1.Inclusion criteria for recurrent miscarriage: two or more consecutive pregnancy loss with the same spouse before 28 weeks of gestation, including biochemical pregnancy; Clinical symptoms include a history of menopause, little or no vaginal bleeding, intrauterine pregnancy indicated by B-ultrasonography, and no germ or centreless tube pulsation.
2.Control group included: normal pregnant women who underwent induced abortion for non-medical reasons within 12w of pregnancy; It matched the gestational age of early RSA patients. Deliver at least one healthy baby at full term; B-ultrasonography showed no abnormal embryo development, with germ and cardiac tube pulsation.

排除标准：

1.复发性流产患者排除标准：月经周期异常；生殖器感染；子宫腔结构异常；抗磷脂综合征；夫妻双方染色体异常及本次流产绒毛染色体核型异常；既往有慢性疾病史者（慢性高血压、糖尿病、肾病、甲状腺疾病、自身免疫性疾病、心血管疾病）。
2.对照组受试者排除标准：月经周期异常；既往有自然流产史；本次妊娠有阴道出血、下腹痛先兆流产症状；生殖器感染；子宫腔结构异常；既往有慢性疾病史者（慢性高血压、糖尿病、肾病、甲状腺疾病、自身免疫性疾病、心血管疾病）。

Exclusion criteria：

1.Exclusion criteria for patients with recurrent abortion: abnormal menstrual cycle; Genital infection; Abnormal uterine cavity structure; Antiphospholipid syndrome; Chromosomal abnormalities of both husband and wife and chromosomal karyotype abnormalities of the aborted villi; Previous history of chronic diseases (chronic hypertension, diabetes, kidney disease, thyroid disease, autoimmune disease, cardiovascular disease).
2.Exclusion criteria of control group: abnormal menstrual cycle; A history of spontaneous abortion; This pregnancy has vaginal bleeding, lower abdominal pain threatened abortion symptoms; Genital infection; Abnormal uterine cavity structure; Previous history of chronic diseases (chronic hypertension, diabetes, kidney disease, thyroid disease, autoimmune disease, cardiovascular disease).
