研究目的：

1.基于本团队对2型糖尿病脾胃湿热证及黄芪四妙汤的前期工作进一步研究，为2型糖尿病脾胃湿热证的生物学基础探索提供表观遗传学参考； 2.观察黄芪四妙汤对2型糖尿病脾胃湿热证差异基因甲基化表达的影响，从DNA甲基化角度探讨黄芪四妙汤治疗2型糖尿病脾胃湿热证的作用机制； 3.通过功能验证及分析，探讨黄芪四妙汤干预2型糖尿病脾胃湿热证DNA甲基化相关的关键基因及通路。  

Objectives of Study：

1. Based on the team's further research on the preliminary work of type 2 diabetes with spleen stomach damp heat syndrome and Huangqi Simiao Decoction, it provides epigenetic reference for the exploration of the biological basis of type 2 diabetes with spleen stomach damp heat syndrome; 2. To observe the effect of Huangqi Simiao Decoction on the expression of differential gene methylation in type 2 diabetes with spleen stomach damp heat syndrome, and to explore the mechanism of Huangqi Simiao Decoction in treating type 2 diabetes with spleen stomach damp heat syndrome from the perspective of DNA methylation; 3. Through functional verification and analysis, explore the key genes and pathways related to DNA methylation in the intervention of Huangqi Simiao Decoction in type 2 diabetes with spleen stomach damp heat syndrome.

药物成份或治疗方案详述：

 

Description for medicine or protocol of treatment in detail：

 

纳入标准：

①符合2型糖尿病的诊断标准； ②经2位副主任中医师以上专家进行辨证，符合2型糖尿病脾胃湿热证的中医诊断； ③年龄18～70岁； ④签署知情同意书； ⑤患者能提供详细的联络方式，无短期迁移计划，愿意配合随访。

Inclusion criteria

① Conform to the diagnostic criteria of type 2 diabetes; ② According to the syndrome differentiation conducted by two experts at or above the level of deputy chief Chinese physician, it is in line with the TCM diagnosis of spleen stomach damp heat syndrome of type 2 diabetes; ③ Age 18-70 years old; ④ Sign an informed consent form; ⑤ The patient is able to provide detailed contact information and has no short-term migration plan, and is willing to cooperate with follow-up.

排除标准：

①六个月内有糖尿病酮症酸中毒、恶性高血压、心肌梗塞、脑血管意外、心衰等危急重症病史者； ②近期内(4周内)有严重感染者； ③合并心、肝、脑、肾和造血系统等严重原发性疾病，癌症、精神病患者； ④妊娠或准备妊娠、哺乳期妇女； ⑤已知对该类药物过敏及严重过敏体质者； ⑥近3个月内参加其他临床药物试验的患者； ⑦对本研究了解不充分、不愿参加，或依从性差者；不合作者(不能配合饮食控制,或不按规定用药而影响疗效判定者)。

Exclusion criteria：

① Those who have a history of critical and serious diseases such as diabetes ketoacidosis, malignant hypertension, myocardial infarction, cerebrovascular accident, heart failure, etc. within six months; ② There are severe infections in the near future (within 4 weeks); ③ Patients with severe primary diseases such as heart, liver, brain, kidney, and hematopoietic system, as well as cancer and mental illness; ④ Pregnant or preparing pregnant or lactating women; ⑤ Individuals who are known to be allergic to or severely allergic to this type of drug; ⑥ Patients who have participated in other clinical drug trials within the past 3 months; ⑦ Those who have insufficient understanding of this study, are unwilling to participate, or have poor compliance; Non cooperative individuals (those who cannot cooperate with dietary control or do not use medication according to regulations, which affects the determination of efficacy).
