研究目的：

比较IVF-PGT与ICSI-PGT的优劣，明确IVF受精是否会导致非整倍体胚胎占比升高。同时，为探索IVF-PGT用于非男性因素不孕夫妇的临床获益及IVF-PGT在临床上的精准应用，提供高质量循证医学的证据。  

Objectives of Study：

In couples with non-male factor infertility, we definite whether the conventional IVF lead to an increase in the proportion of aneuploid embryos by comparing the advantages and disadvantages between IVF-PGT and ICSI-PGT. In addition, to explore the clinical benefits of IVF-PGT in couples with non-male factor infertility and the accurate clinical application of IVF-PGT,the randomised controlled muli-centric trial(RCT) will provide the high-quality evidence-based madicine(EBM).

Description for medicine or protocol of treatment in detail：

 

纳入标准：

受试者必须符合以下所有标准才有资格参与本试验：
（1）符合IVF条件，PGT-A周期患者；
（2）纳入患者的促排取卵后，获卵数≥6枚；
（3）女性年龄40周岁以下，BMI值范围18~30kg/m2；
（4）男方精液指标正常（精液浓度>5×10^6/mL，向前运动精子数≥5×10^6个）；

Inclusion criteria

To be eligible to participate in the trial, subjects must meet all of the following criteria:
(1) Eligible for IVF, PGT-A cycle patients;
(2) At least six oocytes obtain following oocyte retrieval after ovulation induction treatment;
(3) Femal age ≤ 40 years, 18kg/m2 ≥ BMI ≤30kg/m2;
(4) The male semen index was normal (semen concentration >5×10^6/mL,number of progressive sperm ≥5×10^6);

排除标准：

受试者符合下列任一标准则不能参与本试验：
（1）IVF或ICSI禁忌患者：Ⅰ型、Ⅱ型糖尿病患者；未确诊肝病患者或者功能紊乱；肾脏疾病或血清肾功异常；贫血症；有深静脉栓、肺静脉栓或脑血管意外病史；高血压失控或有症状的心脏病；有或疑似宫颈癌病史、子宫内膜癌、乳腺癌；无法解释的阴道出血；
（2）男性因素不孕：严重少精症、弱精子症、畸形精子症；
（3）既往周期受精率低下（受精率<25%）；
（4）供卵和供精患者；
（5）使用冻精患者；

Exclusion criteria：

Participants were excluded from the trial if they meet any of the following criteria:
(1) IVF or ICSI contraindicated patients: patients with type ⅰ and type ⅱ diabetes mellitus; Patients with undiagnosed liver diseases or functional disorders; Kidney disease or abnormal serum renal function; Anemia; A history of deep vein thrombosis, pulmonary vein thrombosis, or cerebrovascular accident; Uncontrolled hypertension or symptomatic heart disease; Have or suspected history of cervical cancer, endometrial cancer, breast cancer; Unexplained vaginal bleeding;
(2) Male factor infertility: severe oligospermia, asthenospermia, teratospermia;
(3) Low fertilization rate in previous cycles (fertilization rate <25%);
(4) Oocytes and sperm donor patients;
(5) Patients using frozen sperm;

随机方法（请说明由何人用什么方法产生随机序列）：

各中心胚胎实验室胚胎操作人员将同一受试者的卵子按照1:1的比例随机分入IVF-PGT试验组和ICSI-PGT对照组。具体的随机分组是将同一受试者的卵丘-卵母细胞复合物收集到一个培养皿中，未剥离卵丘颗粒细胞前在低倍显微镜下将一半COCs分配到培养皿中（I组），另一半COCs分配到另一个培养皿中（Ⅱ组）。打开电子生成的随机表，验证I组的授精方法，另一组自然地接受另一种授精方法。

The embryonic operators are responsble for randomly dividing half of the sibling oocytes to the IVF-PGT trial group and the other half are assigned to the ICSI-PGT control group.An electronically generated randomisation list was opened to verify the insemination method.
