中重度烟草依赖者戒烟前后脑功能、转录组、表观遗传学、代谢组及咽部微生态变化研究。  

Objectives of Study：

To study the changes of brain function, transcriptome, epigenetics, metabolome and pharyngeal microecology in moderate and severe tobacco addicts before and after smoking cessation.

药物成份或治疗方案详述：

 

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.年龄25-40岁男性；
2.吸烟指数>200；
3.尼古丁依赖自测量表评分≥5分；
4.右利手；
5.经医院伦理委员会批准，所有入选病例均填写知情同意书。

Inclusion criteria

1. Males aged 25-40 years;
2. Smoking index > 200;
3. Nicotine dependence self-rating scale score >= 5 points;
4. Right-handed;
5. Approved by the ethics committee of the hospital, all the selected patients filled out the informed consent form.

排除标准：

1.严重呼吸系统、心血管、消化系统器质性疾病；
2.中枢神经系统疾病史；
3.中枢神经相关药品使用史；
4.家族遗传病（如糖尿病病史、高血压病史）；
5.基础精神疾病史；
6.创伤史；
7.服用精神类药物史；
8.近期麻醉史；
9.头部外伤史；
10.酒精或药物滥用史；
11.磁共振检查禁忌者。

Exclusion criteria：

1. Severe respiratory, cardiovascular, and digestive system organic diseases;
2. History of central nervous system disease;
3. History of use of central nervous system-related drugs;
4. Family genetic diseases (such as history of diabetes, history of hypertension);
5. History of underlying mental illness;
6. Trauma history;
7. History of taking psychotropic drugs;
8. Recent history of anesthesia;
9. History of head trauma;
10. History of alcohol or drug abuse;
11. Those who are contraindicated in magnetic resonance examination.

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：

试验者样本采集由昆明医科大学第一附属医院生物样本库及昆明医科大学第一附属医院呼吸科执行，转录组测序由天津诺禾致源检测，代谢组由上海厚基生物科技有限公司检测，咽拭子微生物组学由广州微远基因科技有限公司检测，源数据保存至负责人张剑青处，试验者样本信息录入由昆明医科大学第一附属医院呼吸科采集，生物样本库核对。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：

Subjects'specimen collection are implemented by Biobank and Respiratory and Critical Care of the First Aaffiliated Hospital of Kunming Medical University.Transcriptome sequencing are detected by Tianjin Nuohezhiyuan Biological information technology co.LTD.Shanghai Houji biotechnology co.LTD test the metabolome, and Guangzhou Weiyuan Gene Technologyco.LTD complete the pharyngeal microecology.Stady leader ZhangJianqing preserve the source data,Respiratory and Critical Care of the First Aaffiliated Hospital of Kunming Medical University collect the specimen information record of all of the subjects, then Biobank verify all of then.
