研究目的：

通过利用单细胞RNA测序技术（scRNA-seq），针对1型糖尿病患者，分离外周血单核细胞，从患者体内免疫细胞中找到特定的细胞亚群及信号通路，研究它们在疾病中的参与机制，为1型糖尿病的临床治疗提供新的思路和靶点。  

Objectives of Study：

By using the single-cell RNA sequencing technology (scNA-SEQ), we isolated peripheral blood monocytes from patients with type 1 diabetes, identified specific cell subsets and signal pathways from patients' immune cells, and studied their participation mechanism in the disease, providing new ideas and targets for the clinical treatment of type 1 diabetes.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

（1）发病年龄小于15岁且新诊断糖尿病不超过100天。诊断时时体重处于同年龄组下1/4分位或者BMI处于同年龄组下1/2分位；（2）1型糖尿病相关抗体呈阳性（GAD65，IAA，ICA或者ZnT8）或者起始胰岛素治疗10日内胰岛素自身抗体（IA-2和IA-2B）呈阳性；（3）患者发生过糖尿病酮症或者DKA；（4）空腹C-肽小于200pmol/L；（5）无传染性疾病，无其他代谢性疾病，无其他免疫性疾病。（6）排除继发性糖尿病、心功能衰竭、恶性肿瘤、脑血管意外及精神疾病患者。

Inclusion criteria

(1) The onset age is less than 15 years old and the newly diagnosed diabetes is less than 100 days old. Weight in the lower quartile or BMI in the lower quartile at the time of diagnosis; (2) Positive type 1 diabetes-related antibodies (GAD65, IAA, ICA or ZnT8) or positive insulin autoantibodies (IA-2 and IA-2B) within 10 days of initial insulin treatment; (3) Patients with diabetic ketosis or DKA; (4) fasting c-peptide less than 200pmol/L; (5) No infectious diseases, no other metabolic diseases, no other immune diseases. (6) Patients with secondary diabetes, heart failure, malignant tumor, cerebrovascular accident and mental disease were excluded.

排除标准：

继发性糖尿病、心功能衰竭、恶性肿瘤、脑血管意外及精神疾病患者。
