研究目的：

对免疫兼容性小型猪皮肤临时覆盖烧伤创面的安全性和有效性进行评估。  

Objectives of Study：

To evaluate the safety and efficacy of temporary skin covering of burn wounds in immunocompatible minipigs.

药物成份或治疗方案详述：

 

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.病人或者法律授权代理人书面同意书；
2.年龄大于18岁；
3.男性或者无生育潜力女性；
女性必须是: 天然绝经：定义≥55岁，停经≥1 年或者小于55岁，卵泡刺激激素 (FSH) ≥40.0 IU/L；或者外科绝育包括子宫切除，双侧卵巢切除和或输卵管节扎；
4.总烧伤面积<30% 深II或III度烧伤；
5.烧伤创面需要手术切除；
6.烧伤创面临床需要同种异体皮肤移植桥梁覆盖；
7.先前没有接受同种异体皮肤移植；
8.烧伤创面不位于面部。

Inclusion criteria

1. Written consent from the patient or a legally authorized agent;
2. Aged over 18 years;
3. Men or women with no reproductive potential;
Women must be: Natural menopause: defined as >=55 years old, menopause for >=1 year or less than 55 years old, follicle stimulating hormone (FSH) >=40.0 IU/L; or surgical sterilization including hysterectomy, bilateral oophorectomy and/or tubal ligation;
4. The total burn area is less than 30% of deep II or III degree burns;
5. Burn wounds require surgical resection;
6. Allogeneic skin graft bridge coverage is required in clinical burn wounds;
7. Have not previously received an allogeneic skin graft;
8. The burn wound is not located on the face.

排除标准：

1.哺乳期妇女；
2.艾滋病感染者或者其他研究者认为的危害病人安全的情况；
3.正在使用免疫抑制剂：
（1）抗肿瘤药物；
（2）高剂量激素> 10 mg 强的松龙；
（3）肿瘤坏死因子抑制剂；
（4）钙素抑制剂（环孢菌素，他克莫司等）；
（5）其他抗细胞增生或者免疫抑制剂；
4.青霉素类，氨基糖甙类或者两性霉素过敏史；
5.5年内活动性肿瘤包括需要外科手术，化疗，和或放疗。非转移性基地或鳞状皮肤癌或者原位宫颈癌可以入选；
6.3天前实验临床试验性药物病人；
7.先前接受过猪源或者其他异种组织产品，包括但不限于：戊二醛固定猪或者牛源心脏生物瓣膜；戊二醛固定猪皮产品；
8.BMI > 40 kg/m^2；
9.HbA1c ≥ 7.0%；
10.30天内使用糖皮质激素包括吸入；
11.电击伤或者化学性烧伤；
12.终末期肾病,定义为 MDRD CrCl < 15 mL/min, 或者正在接受长期透析 ；
13.Child-Pugh Score C肝功能病人；
14.3个月内心肌梗塞，需要住院的不稳定心绞痛，需要或者有计划介入（PCI, CABG,颈部或者外周血管再通）治疗的心脑血管疾病 ，短暂缺血发作，需要治疗外周血管疾病；
15.影响烧伤创面血管的外周动，静脉疾病；
16.合并溶血性贫血；
17.研究者认定的长期营养不良；
18.显著肺功能失代偿；
19.皮肤移植治疗期间全身性抗凝药物使用或者INR > 2；
20.治疗前有创面感染；
21.有脓毒血症和或合并有器官功能受损；
22.急性肺损伤；
23.预期寿命短于180天。

Exclusion criteria：

1. Lactating patients;
2. HIV-infected persons or other situations that the researchers believe endanger the patient's safety;
3. Immunosuppressants are being used:
(1) Antitumor drugs;
(2) High-dose hormones > 10 mg prednisolone;
(3) Tumor necrosis factor inhibitors;
(4) Calcin inhibitors (cyclosporine, tacrolimus, etc.);
(5) Other anti-cell proliferation or immunosuppressive agents;
4. History of allergy to penicillin, aminoglycoside or amphotericin;
5. Active tumors within 5 years including the need for surgery, chemotherapy, and/or radiation. Non-metastatic basal or squamous skin cancer or cervical cancer in situ can be included;
6. Patients with clinical trial drugs tested 3 days ago;
7. Previously received porcine-derived or other xenogeneic tissue products, including but not limited to: glutaraldehyde-fixed porcine or bovine-derived heart biological valves; glutaraldehyde-fixed porcine skin products;
8. BMI > 40 kg/m^2;
9. HbA1c >= 7.0%;
10. Glucocorticoid use within 30 days including inhalation;
11. Electrical injury or chemical burn;
12. End-stage renal disease, defined as MDRD CrCl < 15 mL/min, or undergoing long-term dialysis;
13. Child-Pugh Score C liver function patients;
14. Myocardial infarction within 3 months, unstable angina pectoris requiring hospitalization, cardiovascular and cerebrovascular diseases requiring or planned intervention (PCI, CABG, neck or peripheral vascular recanalization), transient ischemic attack, and peripheral vascular disease requiring treatment;
15. Peripheral arterial and venous diseases affecting the blood vessels of burn wounds;
16. Combined with hemolytic anemia;
17. Long-term malnutrition identified by the investigator;
18. Significant decompensation of lung function;
19. Systemic anticoagulant use or INR > 2 during skin graft treatment;
20. Wound infection before treatment;
21. Sepsis and or combined with impaired organ function;
22. Acute lung injury;
23. Life expectancy is less than 180 days.
