研究目的：

1.通过惯性聚焦原理分离肝癌患者外周静脉血中的的CTCs，判断CTCs的数量、亚群与患者预后及治疗反应的关系； 2.利用单细胞全基因组测序技术检测各个患者CTC细胞及对应组织中的突变、拷贝数变异、突变负荷等，运用生物信息学分析方法分析CTC的异质性及其与临床结局的关系； 3.初步探索肝癌循环肿瘤细胞的体外培养方法及细胞特性，以期获得CTCs细胞系，用于揭示肝癌的转移机制。  

Objectives of Study：

1. Isolate CTCs in peripheral venous blood of patients with liver cancer by the principle of inertial focusing, and determine the relationship between the number and subgroup of CTCs and the prognosis and treatment response of patients; 2. Use single-cell whole-genome sequencing technology to detect mutations, copy number variation, mutation load, etc. in CTC cells and corresponding tissues of each patient, and use bioinformatics analysis methods to analyze the heterogeneity of CTCs and their relationship with clinical outcomes; 3. Preliminarily explore the in vitro culture method and cell characteristics of circulating tumor cells of liver cancer, in order to obtain CTCs cell line, which can be used to reveal the metastasis mechanism of liver cancer.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.经过影像学及血清学检查确诊肝癌或者肝良性占位患者；
2.初诊患者，既往未行任何相关治疗；
3.知情同意；
4.年龄：18岁或以上（成人）。

Inclusion criteria

1. Patients diagnosed with liver cancer or benign liver mass after imaging and serological examination;
2. Newly diagnosed patients who have not received any related treatment before;
3. Informed consent;
4. Age: 18 years old or above (adult).

排除标准：

1.合并其他恶性肿瘤；
2.转移性肝脏肿瘤；
3.伴有重大器质性病变。

Exclusion criteria：

1. Combined with other malignant tumors;
2. Metastatic liver tumor;
3. Associated with major organic lesions.
