研究目的: |
1. 主要目的:本课题将结合16S rDNA扩增子测序、非靶向代谢组学手段解析呼吸道菌群在不同转移潜能非小细胞肺癌中的结构与功能特征,筛选与非小细胞肺癌演进相关的主要菌种及代谢物,并利用培养组学及质谱方法进行菌种纯化培养和鉴定。通过体外细胞培养及肿瘤血行转移小鼠模型,研究呼吸道菌群代谢对宿主呼吸道上皮细胞、非小细胞肺癌细胞、局部免疫细胞群的表型影响及机理,从而为非小细胞肺癌病理进化过程中的呼吸道菌群的作用提供新的理论依据,为肺癌的精准防治奠定基础。 2. 次要目的:鉴定不同CT影像表现的特征性呼吸道菌群及代谢物,探索潜在新型肺癌诊断标志物。 |

Objectives of Study: |
1. Main purpose: This project will combine 16S rDNA amplicon sequencing and non-targeted metabolomics to analyze the structural and functional characteristics of respiratory flora in non-small cell lung cancer with different metastatic potential, and to screen for the evolution of non-small cell lung cancer. The main strains and metabolites were purified, cultured and identified by culturomics and mass spectrometry. Through in vitro cell culture and tumor hematogenous metastasis mouse model, the phenotypic effects and mechanisms of respiratory flora metabolism on host respiratory epithelial cells, non-small cell lung cancer cells, and local immune cell populations were studied, to provide a new theoretical basis for the role of respiratory flora in the pathological evolution of non-small cell lung cancer, and lays the foundation for the precise prevention and treatment of lung cancer. 2. Secondary purpose: To identify the characteristic respiratory flora and metabolites in different CT images, and to explore potential new lung cancer diagnostic markers. |

纳入标准: |
1.训练队列纳入临床特征(性别、年龄、吸烟史、基础疾病等)匹配的经病理活检确诊的原发性肺腺癌、肺鳞状细胞癌及肺良性疾病(结核瘤、支气管扩张、错构瘤、真菌感染等)患者,其中早期转移组(T分期≤T2a、N分期≥N1,低分化)、局部侵袭组(局部晚期无淋巴结及远处转移肺癌)(T分期≥T3、N分期=N0,高-中分化)及肺良性疾病组病例各30例。 |

Inclusion criteria |
1. The training cohort included patients with primary lung adenocarcinoma confirmed by pathological biopsy with matching clinical characteristics (sex, age, smoking history, underlying diseases, etc.), patients with lung squamous cell carcinoma and benign lung diseases (tuberculoma, bronchiectasis, hamartoma, fungal infection, etc.), among them, there were 30 cases in the the early metastasis group (T stage<=T2a, N stage>=N1, poorly differentiated), local invasion group (locally advanced lung cancer without lymph node and distant metastasis) (T stage >= T3, N stage = N0, high-moderate differentiation) and benign lung disease group. |
