研究目的：

1.探讨 AMPK 信号通路上基因的 SNP-SNP 互作对子、SNP 集和 SNP 互作网络与 T2DM、CAD 及其共病的关联性。从遗传学角度，为进一步研究 T2DM、CAD 及其共病的发病机制提供科学依据； 2.探讨 AMPK 信号通路上基因的 DNA 甲基化修饰的 CpG-SNP 与 T2DM、CAD 及其共病之间的关联性，评价其对 T2DM、CAD 及其共病罹患风险的贡献大小；同时研究定位克隆 T2DM、CAD 及其共病相关 DNA 甲基化修饰的特异性CpG-SNP，并在细胞水平上进行功能学验证。从表观遗传学角度，为深入研究T2DM、CAD 及其共病的发病机制以及寻找新的分子标志物和药物作用靶点提供理论依据  

Objectives of Study：

1. To explore the association between SNP-SNP interaction pairs, SNP sets and SNP interaction networks of genes in AMPK signaling pathway and T2DM, CAD and its comorbidities. From the perspective of genetics, it provides scientific basis for further study on the pathogenesis of T2DM, CAD and its comorbidities. 2. To explore the association between CpG-SNP modified by DNA methylation of AMPK signaling pathway and T2DM, CAD and its comorbidities, and to evaluate its contribution to the risk of T2DM, CAD and its comorbidities; and to study the specific CpG-SNPs of localized cloning of T2DM, CAD and comorbidities associated with DNA methylation modification, and carry out the functional verification at the cellular level.From the perspective of epigenetics, this study provides a theoretical basis for the further study of the pathogenesis of T2DM, CAD and its comorbidities, as well as the search for new molecular markers and drug action targets

Description for medicine or protocol of treatment in detail：

 

纳入标准：

T2DM 组
(1) 年龄 40 岁-70 岁；
(2) 随机血糖≥11.1 mmol/L，伴有糖尿病症状（高血糖所导致的多饮、多食、多尿、体重下降、皮肤瘙痒、视力模糊等急性代谢紊乱表现），或无糖尿病症状，空腹血糖≥7.0 mmol/L，或葡萄糖负荷后 2 小时血糖≥11.1 mmol/L；
(3) 无 CAD、高血压及其它心脑血管疾病，T1DM，严重肝肾疾病和恶性肿瘤等病史

CAD 组
(1) 年龄 40 岁-70 岁；
(2) 根据冠状动脉造影术(CAG)或 64 层螺旋 CT 冠状动脉成像确定至少一只冠状动脉狭窄≥50%；
(3) 无糖尿病及其它内分泌系统疾病，严重肝肾疾病和恶性肿瘤等病史

共病组
(1) 年龄 40 岁-70 岁；
(2) 同时符合 T2DM 和 CAD 诊断标准的患者；
(3) 无高血压及其它心脑血管疾病，T1DM 及其它内分泌系统疾病，严重肝肾疾病和恶性肿瘤等病史。

对照组
(1) 年龄 40 岁-70 岁；
(2) 无糖尿病、心血管疾病家族遗传史；
(3) 无糖尿病，冠心病、高血压及其它心脑血管疾病，严重肝肾疾病和恶性肿瘤等病史；
(4) 经询问病史、体检、血糖检查及其它检查结果无异常。

Inclusion criteria

T2DM group
(1) The age ranged from 40-70 years old;
(2) Random blood glucose≥11.1 mmol/L, accompanied by diabetes symptoms (hyperglycemia caused by hyperglycemia, polyuria, weight loss, skin pruritus, blurred vision and other acute metabolic disorders), or no diabetes symptoms, fasting blood glucose ≥7 mmol/L, or 2 hours after glucose loading, blood glucose≥11.1 mmol/L;
(3) There was no history of CAD, hypertension, other cardiovascular and cerebrovascular diseases, T1DM, severe liver and kidney diseases and malignant tumor

CAD group
(1) The age ranged from 40-70 years old;
(2) According to coronary angiography (CAG) or 64 slice spiral CT coronary angiography, at least one coronary artery stenosis ≥ 50% was determined;
(3) no diabetes and other endocrine diseases, serious liver and kidney diseases and malignant tumors.

Comorbidity group
(1) The age ranged from 40-70 years old;
(2) Patients who met the diagnostic criteria of T2DM and CAD at the same time;
(3) There was no history of hypertension and other cardiovascular and cerebrovascular diseases, T1DM and other endocrine diseases, serious liver and kidney diseases and malignant tumors.

control group
(1) The age ranged from 40-70 years old;
(2) no family history of diabetes and cardiovascular diseases.
(3) no diabetes, coronary heart disease, hypertension and other cardiovascular and cerebrovascular diseases, serious liver and kidney diseases and malignant tumors.
(4) The results of medical history, physical examination, blood glucose examination and other examinations were normal.

排除标准：

有高血压，严重肝肾疾病和恶性肿瘤等病史
