Objectives of Study：

1. Evaluate the effectiveness of electroacupuncture on non-alcoholic steatohepatitis 2. Evaluate the safety of electroacupuncture on non-alcoholic steatohepatitis

药物成份或治疗方案详述：

 

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.年龄18-65岁男性或女性患者（包含18岁和65岁）；
2.影像学显示脂肪肝（B超或CT或核磁）；
3.NASH确诊或NASH表型诊断
A．NASH确诊：有病史证明在筛选前6个月内或筛选期内经肝脏病理活检具有NASH的组织学证据（NAS评分≥4分，其中炎症和气球样变各至少1分，纤维化水平≤F3），组织学诊断未发现其他慢性肝病且ALT≥1.5×ULN；
B．NASH表型诊断：3个月内间隔大于7天的两次检查ALT≥1.5×ULN和BMI≥25 kg/m2和基线时肝脏脂肪≥8%（MRI-PDFF）。
4.签署知情同意书

Inclusion criteria

1. Male or female patients aged 18-65 (including 18 and 65 years old);
2. Imaging shows fatty liver (B-ultrasound or CT or MRI);
3. NASH diagnosis by biopsy or clinical features
A. NASH diagnosis by biopsy: histological evidence of NASH by liver biopsy within 6 months before screening or during the screening period (NAS score ≥ 4 points, including inflammation and ballooning at least 1 point each, and fibrosis level ≤ F3), No other chronic liver diseases were found in histological diagnosis and ALT≥1.5×ULN;
B. NASH diagnosis by clinical features: two examinations with an interval greater than 7 days within 3 months and BMI≥25 kg/m2 and liver fat ≥8% by MRI-PDFF at baseline.
4. Sign the informed consent

排除标准：

1.通过病史、实验室检测怀疑有其它原因的肝胆疾病，包括但不限于：乙型或丙型肝炎病毒感染、慢性酒精性肝病、药物性肝病、自身免疫性肝炎、原发性胆汁性肝硬化、原发性硬化性胆管炎、Wilson’s病、α1-抗胰蛋白酶缺乏症、肝癌、明显的肝功能异常（ ALT或AST≥5×ULN或TBIL≥ 1.5×ULN）等；
2.筛选前1年内过量饮酒连续3个月或以上（男性平均每天饮用乙醇超过30克，相当于3.75单位酒精，女性超过20克，相当于2.5单位酒精：1单位=啤酒285 mL，或烈酒25mL，或葡萄酒100mL）；
3.有减肥手术史或近期准备行减肥手术者；
4.肝活检提示肝硬化或既往临床诊断为肝硬化；
5.1型糖尿病患者和未受控制的2型糖尿病患者（HbA1c≥9.5%）；
6.筛选前其它临床发现显示任何病情不稳定或未进行治疗的有临床意义的下列疾病（包括但不限于胃肠道、神经、血液、内分泌、肿瘤、肺、免疫、精神疾病）
7.有急性心血管事件发生的病史，或有以下心脑血管病史：冠状动脉成形术、脑卒中、短暂性脑缺血发作、冠心病的患者；
8.正在服用可能引起脂肪变/脂肪性肝炎的药物（包括胺腆酮、甲氨碟呤、激素、四环素、他莫西芬、类固醇、丙戊酸等）；
9.随机分组前90天服用降糖药GLP-1、噻唑烷二酮类胰岛素增敏剂相关药物、减重药(包括并不限于西布曲明、奥利司他)和临床显示可能有改善脂肪性肝炎的新药(包括并不限于利拉鲁肽、奥贝胆酸、Elafibranor、维生素 E 等);
10.在参加试验前1个月内进行过针灸治疗的患者；
11.妊娠和哺乳期女性或近期有生育需求未避孕的女性；
12.有MRI扫描禁忌的患者（携带金属填充物的假体，空间狭小紧张综合征等）；
13.随机入组前8周体重变化超过5%的患者；
14.依研究者判断不适于参加研究者。

Exclusion criteria：

1. Other hepatobiliary diseases, including but not limited to: hepatitis B or C virus infection, chronic alcoholic liver disease, drug-induced liver disease, autoimmune hepatitis, primary biliary liver Sclerosis, primary sclerosing cholangitis, Wilson's disease, α1-antitrypsin deficiency, liver cancer, obvious liver function abnormalities (ALT or AST≥5×ULN or TBIL≥1.5×ULN), etc.;
2. Excessive drinking for 3 consecutive months or more in the 1 year before screening (the average daily consumption of ethanol for men is more than 30 grams, which is equivalent to 3.75 units of alcohol, and for women, more than 20 grams, which is equivalent to 2.5 units of alcohol: 1 unit = 285 mL of beer, or strong 25mL wine, or 100mL wine);
3. The history of bariatric surgery or the plan to bariatric surgery in the near future;
4. Liver biopsy indicates liver cirrhosis or a previous clinical diagnosis of liver cirrhosis;
5. Type 1 diabetes patients and uncontrolled type 2 diabetes patients (HbA1c≥9.5%);
6. Other diseases that are unstable or untreated (including but not limited to the gastrointestinal tract, nerve, blood, endocrine, tumor, lung, immune, mental diseases)
7. Patients with a history of acute cardiovascular events, or a history of the following cardiovascular and cerebrovascular diseases: coronary angioplasty, stroke, transient ischemic attack, coronary heart disease;
8. Taking drugs that may cause steatosis/steatohepatitis (including amoxicillin, methotrexate, hormones, tetracycline, tamoxifen, steroids, valproic acid, etc.);
9. 90 days before randomization, taking hypoglycemic drugs GLP-1, thiazolidinedione insulin sensitizer-related drugs, weight loss drugs (including but not limited to sibutramine, orlistat) and clinically shown to improve steatohepatitis (including but not limited to liraglutide, obeticholic acid, Elafibranor, vitamin E, etc.);
10. Patients who have undergone acupuncture treatment within 1 month before participating in the trial;
11. Pregnant and lactating women or women who have recently had childbirth need without contraception;
12. Patients with contraindications for MRI scanning (prostheses with metal fillers, tight space syndrome, etc.);
13. Patients with a weight change of more than 5% in the 8 weeks before randomization;
14. According to the researcher's judgment, it is not suitable to participate in the research.

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：

1.数据采集：严格遵守研究方案，以保证按照GCP执行，如实记录原始病例，并将相关内容及时、如实、详细、认真填写eCRF。 2.数据管理：采用艾莎医学公司提供的EDC系统采集数据，在临床研究和数据处理阶段均有相应的数据管理措施。
