研究目的：

以PDE为切入点，建立维生素B6相关性癫痫的临床规范化筛选、诊断、治疗及预后评估流程。以PDE为研究对象，建立维生素B6及特殊饮食治疗的规范化流程并进行有效性和安全性评估。印证这一新疗法对智力运动发育情况和体内生化标志物的影响，探索出更为合适的特殊饮食治疗流程及方案，改善PDE的远期认知预后。通过以PDE特殊饮食治疗为切入点，为进一步开展其他维生素B6相关性癫痫的特殊治疗方法开拓思路和方法。  

Objectives of Study：

With PDE as the entry point, the clinical standardized screening, diagnosis, treatment and prognosis assessment of vitamin B6-related epilepsy were established. Taking PDE as the research object, the standardized procedures of vitamin B6 and special dietary therapy were established and the efficacy and safety were evaluated. To verify the influence of this new therapy on the development of mental motor and biochemical markers in vivo, explore a more appropriate special diet treatment process and program, and improve the long-term cognitive prognosis of PDE. Through the PDE special diet treatment as the entry point, for the further development of other vitamin B6-related epilepsy special treatment ideas and methods.

药物成份或治疗方案详述：

 

Description for medicine or protocol of treatment in detail：

 

纳入标准：

（1）符合PDE的临床诊断标准；（2）经遗传学检测确诊：确定携带ALDH7A1基因突变（纯合或复合杂合突变）。
上述被试者的家长或监护人知情，签署知情同意书。

Inclusion criteria

(1) Meet the clinical diagnostic criteria of PDE; (2) Confirmed by genetic testing: confirmed to carry ALDH7A1 gene mutation (homozygous or complex heterozygous mutation).
The parents or guardians of the subjects were informed and signed the informed consent.

排除标准：

（1）不能配合接受限制赖氨酸饮食治疗者，或出现严重不良反应（营养，神经或其他）；（2）患有肝功能异常等疾病，可能影响生化标志物水平者；（3）维生素B6单一治疗后癫痫发作停止，并建立正常的行为和发育者；（4）由于视觉和听觉感官缺陷引起发展迟缓者。

Exclusion criteria：

(1) those who cannot cooperate with the treatment of lysine-restricted diet, or have serious adverse reactions (nutritional, neurological or other); (2) patients with abnormal liver function and other diseases that may affect the level of biochemical markers; (3) after vitamin B6 monotherapy, the seizures stopped and normal behavior and development were established; (4) Developmental delay caused by visual and auditory sensory defects.

将确诊的PDE患儿按照完全随机化分组的方法分为饮食干预组和对照组，每组分别20-25例。具体方法：①编号：首先将全部入组患者按照就诊次序从1到N排序编号；②获取随机数字：从随机数表中任意一个数开始，沿同一方向顺序获取每个试验单位一个随机数字；③求余数：随机数除以编号求余数，若整除则余数取组数；④分组：按余数的奇偶分为1、2两组；⑤调整：假如共有n例待调整，需要从中抽取1例，便继续抄一个随机数，除以n后将得到的余数作为所抽试验单位的序号（若整除则余数为n）。
