研究目的：

本项目将通过已有的多功能免疫脂质磁球（EpCAM、Vimentin等多位点）分离技术将循环肿瘤细胞（CTC）进行分离，在免疫荧光技术的基础上，结合加权引导滤波的Retinex细胞图像增强算法及轻量级卷积神经网络的CTC智能识别方法，针对CTC图像存在灰度不均匀、低对比度以及边缘模糊等特点，利用深度学习等人工智能方法，进行分割与识别，对胃癌患者血液中的CTC进行靶向分离及精准鉴定，实现对CTC的精准识别，建立基于CTC的早期胃癌精准诊断模型；同时，在获得的CTC单细胞基础上，结合CTC单细胞的免疫组化/Fish信息，及CTC单细胞的基因突变和基因组学测试数据，整合临床医疗数据（病理、肿瘤分级分期、化疗敏感性、耐药性、放疗敏感性、预后预测等）、专业文献等，再将生物信息和临床信息结合起来，基于CTC液体活检的大数据开展相关科学研究，构建智能筛查的胃癌精准诊疗评估系统，并最终对患者进行精准的个体化诊疗评估分析，实现对早期胃癌的精准诊断及胃癌患者临床疗效的精准评估。  

Objectives of Study：

This project will separate circulating tumor cells (CTC) through the existing multi-functional immunolipid magnetic sphere (EpCAM, Vimentin, etc.) separation technology. On the basis of immunofluorescence technology, combined with weighted guided filter Retinex cells Image enhancement algorithm and CTC intelligent recognition method of lightweight convolutional neural network. Aiming at the characteristics of CTC images such as uneven gray scale, low contrast and blurred edges, using artificial intelligence methods such as deep learning to segment and recognize patients with gastric cancer Targeted separation and precise identification of CTCs in the blood, to achieve precise identification of CTCs, to establish a CTC-based accurate diagnosis model for early gastric cancer; at the same time, on the basis of the obtained CTC single cells, combined with CTC single cell immunohistochemistry/Fish Information, gene mutations and genomics test data of CTC single cells, integration of clinical medical data (pathology, tumor grade and staging, chemotherapy sensitivity, drug resistance, radiotherapy sensitivity, prognosis prediction, etc.), professional literature, etc., and then biological Information and clinical information are combined, and relevant scientific research is carried out based on the big data of CTC liquid biopsy, to build an intelligent screening and accurate diagnosis and treatment evaluation system for gastric cancer, and finally to conduct accurate and individualized diagnosis and treatment evaluation and analysis of patients to achieve accurate diagnosis of early gastric cancer And accurate assessment of clinical efficacy in patients with gastric cancer.

纳入标准：

a. 经多学科讨论后认定的影像学诊断的胃癌患者；
b. 年龄≥18岁；
c. 经CT或MRI检查有可测量或可评估的肿瘤病灶（根据RECIST 1.1标准）；
d. KPS评分大于60；
h. 签署知情同意书。

Inclusion criteria

a. Patients with gastric cancer confirmed by imaging diagnosis after multidisciplinary discussion;
b. Age ≥ 18 years old;
c. There are measurable or evaluable tumor lesions by CT or MRI (according to RECIST 1.1 standard);
d. KPS score is greater than 60;
h. Sign the informed consent form.

排除标准：

a. 无胃癌的病理学证据；
b. 既往患其它恶性肿瘤的患者；
c. 感染活动期或其它可能干扰本项目检测结果的严重感染；
d. 怀孕或哺乳期女性；
e. 正接受校正监视或监护的个体；
f. 其它可能干扰本项目检测结果的状况。

Exclusion criteria：

a. No pathological evidence of gastric cancer;
b. Patients suffering from other malignant tumors in the past;
c. Active infection period or other serious infections that may interfere with the test results of this project;
d. Pregnant or breastfeeding women;
e. Individuals undergoing correction monitoring or guardianship;
f. Other conditions that may interfere with the test results of this project.
