本研究旨在分析PBC患者肠道菌群分布特征与骨质代谢紊乱的相关性,通过粪便16S高通量测序及代谢组学分析筛选出与骨代谢紊乱相关的致病菌及其代谢产物;通过血浆样本转录组学分析筛选出特异性靶基因,为进一步开展PBC骨病的分子机制研究提供坚实的临床依据,为PBC骨病患者寻找有效的治疗靶点。 |

Objectives of Study: |
This study aims to analyze the correlation between the distribution characteristics of the intestinal flora of PBC patients and bone metabolism disorders, and screen out pathogenic bacteria and their metabolites related to bone metabolism disorders through 16S high-throughput sequencing and metabonomics analysis of stool; Transcriptomics analysis of plasma samples is used to screen out specific target genes, which will provide a solid clinical basis for further research on the molecular mechanism of PBC bone disease and find effective therapeutic targets for patients with PBC bone disease. |

数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC: |
? 采用纸质化数据管理模式,在试验启动之前对试验参加人员进行相关培训。 ? 各研究中心临床研究者应指定数据录入员,录入应及时、准确并签名。 ? 对CRF中存在的疑问。质控员将随时提出疑问,研究者应尽快给予解答,修改错误数据。 ? 在每一位观察对象完成试验并经质控员审核无误后,由数据管理员对数据进行锁定,直至完成最后一位观察对象的数据的锁定。数据全部锁定后,由数据管理员将其导入到指定数据库,交统计人员进行统计分析。 ? 主要研究者根据统计报告写出临床研究总结报告。 |

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture: |
? Adopt paper-based data management mode, and conduct relevant training for test participants before the start of the test. ? Clinical investigators in each research center should appoint data entry personnel, and entry should be timely, accurate and signed. ? Questions in CRF. The quality controller will ask questions at any time, and the researcher should give answers as soon as possible and modify the wrong data. ? After each observation object completes the test and is checked by the quality controller, the data administrator will lock the data until the data of the last observation object is locked. After all the data is locked, the data administrator will import it into the designated database and submit it to the statistician for statistical analysis. ? The main investigator writes a clinical research summary report based on the statistical report. |
