青少年特发性脊柱侧凸(AIS)的发病机制未明,研究发现脊柱前柱过度生长(RASO)是AIS发生的关键因素.近年来RASO成为AIS病因学研究的前沿热点,但其发生机制尚不明确.申请人发现AIS患者血清松弛素3(RLN3)水平显著高于正常;RLN3可通过降低韧带成纤维细胞活性改变韧带胶原含量;RLN3受体RXFP3拮抗剂可显著抑制RLN3的抗纤维化作用.据此推测,RLN3可能是AIS发病的关键分子,其可通过受体RXFP3调控韧带抗纤维化引起胶原异常导致脊柱前方韧带松弛,后者可诱发脊柱前柱过度生长促进AIS发病.本研究拟通过多中心临床样本探寻RLN3作为AIS诊断预测指标的可行性;通过AIS疾病模型筛选具有治疗作用的RLN3-RXFP3轴拮抗剂,为AIS的诊疗和干预提出新思路。 |

Objectives of Study: |
The pathogenesis of adolescent idiopathic scoliosis (AIS) is unknown. Studies have found that overgrowth of the anterior spine (RASO) is a key factor in the occurrence of AIS. In recent years, RASO has become a frontier hot spot in the etiology of AIS, but its mechanism is not yet clear. Clear. The applicant found that serum relaxin 3 (RLN3) levels in AIS patients were significantly higher than normal; RLN3 can change ligament collagen content by reducing the activity of ligament fibroblasts; RLN3 receptor RXFP3 antagonists can significantly inhibit the anti-fibrotic effect of RLN3. Based on this speculation, RLN3 may be a key molecule in the pathogenesis of AIS. It can regulate ligament anti-fibrosis and cause collagen abnormalities through the receptor RXFP3 to relax the anterior spine ligaments, which can induce excessive growth of the anterior spine and promote the onset of AIS. This study is proposed to be adopted. Multi-center clinical samples explore the feasibility of RLN3 as a diagnostic predictor of AIS; screening therapeutic RLN3-RXFP3 axis antagonists through AIS disease models, and propose new ideas for AIS diagnosis, treatment and intervention. |

确诊为神经肌肉型脊柱畸形、先天性脊柱畸形等非特发性脊柱侧凸,男性,年龄超过18岁或小于12岁,RISSER征大于IV° |
