1. 明确 SIRT5-SREBF1-SCD1 轴在 ccRCC 组织中的表达特征及其临床预后价值 利用回顾性收集的临床病理样本(FFPE),通过免疫组化(IHC)检测 SIRT5、SREBF1、SCD1 在肾透明细胞癌及癌旁组织中的表达差异;结合患者的临床随访数据,分析该轴的异常表达与临床病理参数(如分级、分期)及生存结局(OS、PFI)的相关性,评估其作为预后标志物的潜力。 2. 探索 SIRT5-SREBF1-SCD1 轴相关的转录组学特征及潜在调控网络 选取具有代表性的临床样本进行全转录组测序(RNA-seq),分析 SIRT5 高/低表达组之间的差异基因(DEGs);重点阐明该轴与脂质代谢相关基因集、缺氧通路及下游信号网络的共表达关系,从转录水平揭示其调控 ccRCC 恶性进展的潜在分子基础。 3. 验证 SIRT5-SREBF1-SCD1 轴在 ccRCC 发生发展中的生物学功能(辅助验证) 在体外细胞模型及动物移植瘤模型中,通过基因敲除或过表达手段,验证该轴对 ccRCC 细胞增殖、侵袭、转移及血管生成能力的调控作用,确认其在组织样本中观察到的临床现象具有生物学功能基础。 |

Objectives of Study: |
1. Clarify the expression characteristics of the SIRT5-SREBF1-SCD1 axis in ccRCC tissues and its clinical prognostic value. Retrospectively collected clinicopathological samples (FFPE) are used to detect the differential expression of SIRT5, SREBF1 and SCD1 in clear cell renal cell carcinoma and adjacent normal tissues via immunohistochemistry (IHC). Combined with patients' clinical follow-up data, the correlation between the abnormal expression of this axis and clinicopathological parameters (such as grading and staging) as well as survival outcomes (OS, PFI) is analyzed to evaluate its potential as a prognostic biomarker. 2. Explore the transcriptomic characteristics and potential regulatory networks associated with the SIRT5-SREBF1-SCD1 axis. Representative clinical samples are selected for whole-transcriptome sequencing (RNA-seq) to analyze differentially expressed genes (DEGs) between the SIRT5 high-expression and low-expression groups. Emphatically elucidate the co-expression relationship between this axis and lipid metabolism-related gene sets, hypoxia pathways and downstream signaling networks, and reveal the potential molecular mechanism by which it regulates the malignant progression of ccRCC at the transcriptional level. 3. Verify the biological functions of the SIRT5-SREBF1-SCD1 axis in the occurrence and progression of ccRCC (auxiliary verification). 纳入标准: |
1. 年龄 >= 18 岁,性别不限; 2. 在本院行根治性肾切除术或肾部分切除术,术后病理组织学明确诊断为肾透明细胞癌(ccRCC); 3. 临床病理资料完整可溯源(包括患者年龄、性别、肿瘤大小、Fuhrman 分级、TNM 分期及生存随访数据); 4. 病理科留存有足量的福尔马林固定石蜡包埋(FFPE)组织,且样本保存状况良好,满足免疫组化及 RNA 提取的质量要求。 |

Inclusion criteria |
1. Age ≥ 18 years, any gender; 2. Underwent radical nephrectomy or partial nephrectomy at our hospital, with postoperative pathological histology confirming clear cell renal cell carcinoma (ccRCC); 3. Complete clinical and pathological data traceable (including patient age, gender, tumor size, Fuhrman grade, TNM stage, and survival follow-up data); 4. Sufficient formalin-fixed paraffin-embedded (FFPE) tissue retained in the pathology department, with good preservation conditions, meeting the quality requirements for immunohistochemistry and RNA extraction. |

排除标准: |
1. 病理诊断为非透明细胞癌(如乳头状肾细胞癌、嫌色细胞癌等)或良性肿瘤; 2. 术前接受过新辅助化疗、放疗、免疫治疗(如 PD-1/PD-L1 抑制剂)或靶向治疗者(避免治疗对代谢及基因表达的干扰); 3. 合并其他系统的恶性肿瘤病史者; 4. 样本发生严重自溶、霉变,或经评估 FFPE 样本存放时间过久导致 RNA 降解严重,无法满足转录组测序质控要求者。 |

Exclusion criteria: |
1. Pathological diagnosis of non-clear cell carcinoma (such as papillary renal cell carcinoma, chromophobe carcinoma, etc.) or benign tumor; 2. Patients who have received neoadjuvant chemotherapy, radiotherapy, immunotherapy (such as PD-1/PD-L1 inhibitors), or targeted therapy before surgery (to avoid interference of treatments on metabolism and gene expression); 3. Patients with a history of malignant tumors in other systems; 4. Samples that have undergone severe autolysis or mold contamination, or FFPE samples evaluated to have been stored for too long resulting in severe RNA degradation, failing to meet transcriptome sequencing quality control requirements. |
