研究目的：

探讨SPP1是否通过CD44-PI3K通路诱导铁死亡并促进黄韧带成纤维细胞纤维化，为黄韧带肥厚机制及潜在干预靶点提供依据。  

Objectives of Study：

To explore whether SPP1 induces ferroptosis and promotes fibrosis of ligamentum flavum fibroblasts through the CD44-PI3K signaling pathway, so as to provide a basis for the mechanism of ligamentum flavum hypertrophy and potential intervention targets.

药物成份或治疗方案详述：

 

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.经影像学（MRI/CT）及临床诊断明确为腰椎管狭窄症（LSS）伴黄韧带肥厚的患者； 2.手术中切除黄韧带组织并可获得足量标本，用于细胞分离培养或组织学检测； 3.患者年龄 40-60 岁 （包括两端）； 4.无严重感染或系统性疾病影响实验结果； 5.患者知情同意并签署知情同意书。

Inclusion criteria

1.Patients who have been diagnosed with lumbar spinal stenosis (LSS) accompanied by thickening of the ligamentum flavum through imaging (MRI/CT) and clinical diagnosis; 2.During the operation, ligamentum flavum tissue is removed and sufficient specimens can be obtained for cell isolation and culture or histological detection; 3. Patients aged 40-60 years (inclusive); 4. No severe infection or systemic diseases affecting experimental results; 5. Patients are informed and have signed the informed consent form.

排除标准：

1.合并脊柱肿瘤、结核、化脓性感染等疾病者； 2.合并类风湿关节炎、强直性脊柱炎、系统性红斑狼疮等自身免疫性疾病者； 3.合并严重代谢性骨病（如甲状旁腺功能异常、骨软化等）或长期影响骨/韧带代谢的疾病； 4.有明显脊柱外伤史或既往腰椎手术史； 5.长期使用激素、免疫抑制剂或抗肿瘤药物者。

Exclusion criteria：

1.Those with concurrent diseases such as spinal tumors, tuberculosis, and purulent infections; 2.Patients complicated with autoimmune diseases such as rheumatoid arthritis, ankylosing spondylitis and systemic lupus erythematosus; 3. Concurrent severe metabolic bone diseases (such as parathyroid dysfunction, osteomalacia, etc.) or diseases that have long-term effects on bone/ligament metabolism; 4. History of significant spinal trauma or previous lumbar surgery; 5. Long-term use of steroids, immunosuppressants, or antitumor drugs.

This research is an in vitro cell experiment. All the relevant index data of the experiment were uniformly collected and recorded on-site by our research group in our laboratory.
