研究目的：

验证罗伊氏乳杆菌PB-W1和其自然突变株P1D42-6在人体消化道内定植能力的差异，为筛选具有人体定植潜力的益生菌菌株提供理论依据。  

Objectives of Study：

To compare the colonization capacity of Lactobacillus reuteri PB-W1 and its natural mutant P1D42-6 in the human gastrointestinal tract, thereby providing theoretical basis for screening effective probiotic strains with persistent colonization potential in human.

纳入标准：

招募年龄为18–45岁、体质指数（BMI）介于18.5–28 kg/m2之间的健康受试者。

Inclusion criteria

Healthy volunteers (n ≥ 10), aged 18-45 years with a body mass index (BMI) between 18.5 and 28 kg/m2 will be recruited.

排除标准：

1. 具有长期胃肠道疾病（如炎症性肠病、乳糜泻）、糖尿病、心血管疾病、癌症或其他炎症性疾病病史者； 2. 长期服用泻药、降糖药、降压药、降脂药或抗炎药者； 3. 妊娠试验呈阳性（由筛查访视中的尿样检测确定）、或自述处于哺乳期或绝经期的女性； 4. 排便频率少于每日一次者； 5. 每周饮酒超过7次、吸烟（包括咀嚼烟草）或使用非法药物及大麻者； 6. 每周剧烈运动时间超过5小时者； 7. 对任何食物或试验饮食成分存在过敏或不耐受者； 8. 素食者或严格素食者； 9. 在开始试验前3个月内使用过抗生素者； 10. 在开始试验前使用过益生菌、益生元、膳食纤维、omega-3脂肪酸或草本补充剂者（若受试者使用过上述补充剂但符合其他条件，可选择进行为期四周的洗脱期后再参与研究）； 11. 粪便中罗伊氏乳杆菌（Lactobacillus reuteri）细胞含量超过10^4个/克。在实验进行期间，受试者不应服用抗生素和补充益生菌产品，不建议食用酸奶及发酵制品。

Exclusion criteria：

Exclusion criteria are as follows: (1) History of chronic gastrointestinal disorders (e.g., inflammatory bowel disease, celiac disease), diabetes, cardiovascular disease, cancer, or other systemic inflammatory conditions. (2) Long-term use of medications, including laxatives, hypoglycemics, antihypertensives, lipid-lowering agents, or anti-inflammatories. (3) Pregnancy (confirmed by urinalysis at screening), lactation, or postmenopausal status. (4) Defecation frequency of less than once per day. (5) Excessive alcohol consumption (>7 drinks/week), tobacco use, or use of illicit drugs. (6) High levels of vigorous exercise (>5 hours/week). (7) Known allergies or intolerances to any study product components. (8) Adherence to a vegetarian or vegan diet. (9) Antibiotic use within the 3 months preceding the study. (10) Recent use of probiotics, prebiotics, or specific dietary supplements (a 4-week washout period may be permitted for otherwise eligible candidates). Additionally, potential subjects will be screened, and those with baseline fecal L. reuteri levels exceeding 10? CFU/g will be excluded.
