研究目的:

本研究旨在探讨 4 周呼吸肌训练对久坐健康成年人认知功能的影响,并进一步评估其对脑功能活动及呼吸功能指标的改变。同时,通过代谢组学分析探索呼吸训练改善认知功能的潜在生物学机制,为呼吸训练作为一种可行的非药物干预策略提供科学依据。  

Objectives of Study:

This study aims to investigate the effects of a 4-week inspiratory muscle training program on cognitive function in sedentary healthy adults. In addition, the study will evaluate changes in brain functional activity and pulmonary function associated with the intervention. Metabolomic analysis will also be conducted to explore potential biological mechanisms underlying the cognitive improvements induced by respiratory training.

Description for medicine or protocol of treatment in detail:

 

纳入标准:

1.年龄 18–45 岁 的成年人。 2.近 3 个月体重变化不超过 5%。 3.日均久坐或静止时间 ≥8 小时,且中高强度身体活动(MVPA) <150 分钟/周。 4.无明确心血管、呼吸系统、神经系统或精神系统疾病。 5.近期无急性呼吸道感染,且未接受系统性呼吸肌训练或规律运动训练。 6.不吸烟或已戒烟 ≥6 个月。 7.能理解研究内容并 自愿签署知情同意书,能够按要求完成训练与评估。

Inclusion criteria

1.Adults aged 18–45 years. 2.Body weight change less than 5% within the past 3 months. 3.Sedentary behavior ≥8 hours per day, with moderate-to-vigorous physical activity (MVPA) <150 minutes per week. 4.No diagnosed cardiovascular, respiratory, neurological, or psychiatric disorders. 5.No recent acute respiratory infection and no participation in structured respiratory muscle training or regular exercise programs. 6.Non-smokers or individuals who have quit smoking for at least 6 months. 7.Able to understand the study procedures, provide written informed consent, and comply with the training and assessment protocol.

排除标准:

1.既往或目前存在严重心肺疾病、神经系统疾病或骨关节疾病。 2.正在使用或近期调整可能影响呼吸功能或认知功能的药物。 3.存在明显认知功能障碍或精神疾病。 4.研究期间无法保证依从性或预计无法完成研究流程的受试者。

Exclusion criteria:

1.History or presence of serious cardiopulmonary, neurological, or musculoskeletal diseases. 2.Current use or recent adjustment of medications that may affect respiratory function or cognitive performance. 3.Presence of clinically significant cognitive impairment or psychiatric disorders. 4.Individuals who are unable to comply with the study protocol or complete the intervention and assessments during the study period.

Randomization Procedure (please state who generates the random number sequence and by what method):

Allocation concealment will be performed using the opaque, sealed, sequentially numbered envelope method. Randomization will be performed using computer-generated random numbers with a 1:1:1 allocation ratio. The randomization sequence will be generated in advance by an independent researcher who is not involved in the study implementation.

本研究采用单盲设计。受试者根据随机分组接受不同干预措施,但认知功能评估、脑功能检测及数据统计分析的评估人员均不知晓受试者的分组情况,以减少评估偏倚。

Blinding:

This study adopts a single-blind design. Participants will receive different interventions according to their randomized allocation, while outcome assessors and data analysts will be blinded to group assignment to minimize assessment bias.

数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC:

所有受试者的基本信息、干预情况及各项评估指标均由经过培训的研究人员统一记录。数据由两名研究人员独立录入并进行交叉核对,以确保数据准确性。所有数据将进行匿名化处理并存储于受密码保护的计算机系统中,仅研究团队成员可访问。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture:

Trained researchers will collect and record participants' baseline information, intervention details, and outcome measures. All data will be independently entered and cross-checked by two researchers to ensure accuracy. The dataset will be anonymized and stored on password-protected computers, accessible only to authorized research personnel.
