研究目的:

评估多参数流式细胞术(MFC)检测骨髓增殖性肿瘤(MPN)患者初诊时骨髓中白血病干细胞(LSC)水平在预测白血病转化中的预后价值,以白血病转化(发生/未发生)及临床预后为主要结局指标,通过多因素Cox回归分析确定CD34+CD38-cooktail+LSC及其异常免疫表型亚群(如CD34+CD38-cooktail+等)是否可区分各MPN亚型以及是否为MPN患者白血病转化的独立危险因素。  

Objectives of Study:

To evaluate the prognostic value of detecting leukemia stem cell (LSC) levels in bone marrow at diagnosis of myeloproliferative neoplasms (MPN) using multiparameter flow cytometry (MFC) for predicting leukemic transformation. The primary outcome measures are leukemic transformation (occurred/not occurred) and clinical prognosis. Multivariable Cox regression analysis will be performed to determine whether CD34+CD38?cocktail+ LSCs and their aberrant immunophenotypic subsets (e.g., CD34+CD38?cocktail+) can distinguish among MPN subtypes and serve as independent risk factors for leukemic transformation in MPN patients.

纳入标准:

1.初治骨髓增殖性肿瘤(MPN)患者; 2.年龄18-70岁; 3.所有入组患者均需签署知情同意书;

Inclusion criteria

1. Patients with newly diagnosed myeloproliferative neoplasms (MPN); 2. Aged 18 to 70 years; 3. All enrolled patients must provide written informed consent.

排除标准:

1.慢性粒细胞白血病患者; 2.患者缺乏依从性。

Exclusion criteria:

1.Patients with chronic myeloid leukemia (CML); 2.Patients who are non-compliant or have poor compliance.
