研究目的：

作为肿瘤微环境中最主要的成分，癌相关成纤维细胞（CAFs）在恶性肿瘤的发生发展中起到了重要作用。在前期研究中，我们基于COL5A3、FN1、POSTN和RARES2基因构建了CAFs风险特征模型，该模型能准确预测乳腺癌患者的预后和免疫治疗反应，并且我们在体外证实POSTN促进了肿瘤细胞迁移和侵袭能力。有研究报道了POSTN在诱导巨噬细胞M2极化促进肿瘤免疫逃避方面的作用，但POSTN与CD8 T细胞的关系尚未明确。在这项研究中，我们将进一步研究CAFs源性 POSTN对乳腺癌的促癌效应以及对CD8 T细胞浸润和活化的影响及其机制，并从多维度分析POSTN在诱导免疫抑制微环境形成导致肿瘤发生发展的具体分子机制。  

Objectives of Study：

As the most important component of the tumor microenvironment, cancer associated fibroblasts (CAFs) play a crucial role in the occurrence and development of malignant tumors. In the previous study, we constructed a risk characteristic model of CAFs based on COL5A3, FN1, POSTN and RARES2 genes. This model can accurately predict the prognosis and immunotherapeutic response of breast cancer patients, and we confirmed that POSTN promotes tumor cell migration and invasion in vitro. There are studies reporting the role of POSTN in inducing macrophage M2 polarization and promoting tumor immune evasion, but the relationship between POSTN and CD8 T cells is not yet clear. In this study, we will further study the cancer promoting effect of CAFs derived POSTN on breast cancer and its impact on CD8 T cell infiltration and activation and its mechanism, and analyze the specific molecular mechanism of POSTN in inducing the formation of immunosuppressive microenvironment to lead to tumor genesis and development from multiple dimensions.

1.原发性乳腺癌患者48名；
2.另需健康成人12名；

Inclusion criteria

1. 48 patients with primary breast cance; 2. 12 healthy adults;

排除标准：

1.无其他原发病灶的肿瘤； 2.健康成人需排除肿瘤、传染病、炎症等疾病；

Exclusion criteria：

1.Tumors without other primary lesions;
2.Healthy adults need to exclude diseases such as tumors, infectious diseases, and inflammation;
