1.绘制转录活化HBV整合区域免疫细胞图谱:运用ST技术结合单细胞测序/scTCR-seq/scATAC-seq,绘制高精确度的HBV整合区域免疫细胞图谱,明确免疫浸润的种类、状态、克隆性以及空间互作网络 2.明确HBV整合区域驱动免疫细胞的机制:探讨HBV整合驱动免疫细胞浸润的关键因素(HBV整合表达的HBsAg、HBV整合区域宿主和病毒DNA的表观修作用、HBV整合断点免疫相关基因) 3.验证最常见的HBV整合断点免疫基因功能(HBV-APOA1):进行动物实验,明确HBV-APOA1嵌合体对局部免疫信号分子表达、巨噬细胞的趋化作用以及病毒控制作用 |

Objectives of Study: |
1. Mapping the immune cell atlas of transcriptionally active HBV integration regions: Using Spatial Transcriptomics (ST) technology combined with single-cell sequencing/scTCR-seq/scATAC-seq, construct a high-precision immune cell atlas of HBV integration regions, and clarify the types, states, clonality, and spatial interaction networks of immune infiltration. 2.Elucidating the mechanisms by which HBV integration regions drive immune cells: Investigate the key factors underlying HBV integration-driven immune cell infiltration (including HBsAg expressed by HBV integration, epigenetic modifications of host and viral DNA in HBV integration regions, and immune-related genes at HBV integration breakpoints). 3. Validating the function of the most common immune gene at HBV integration breakpoints (HBV-APOA1): Conduct animal experiments to clarify the effects of the HBV-APOA1 chimera on the expression of local immune signaling molecules, the chemotaxis of macrophages, and viral control. |

Inclusion criteria |
1.Signed informed consent prior to enrollment and ability to complete the study in accordance with the research protocol; 2.Patients with HBV infection undergoing liver resection; 3.Patients with HBV infection undergoing liver biopsy. |

排除标准: |
1.明确诊断为肝癌、肝转移瘤患者; 2.合并HIV、HCV患者; 3.合并自身免疫性疾病; 4.孕妇. |

|
Exclusion criteria: |
1.Patients with a definite diagnosis of liver cancer or liver metastases; 2.Patients co-infected with HIV or HCV; 3.Patients with comorbid autoimmune diseases; 4.Pregnant women. |
