研究目的：

1. 通过多组学整合分析系统鉴定膀胱癌 CAFs 的功能亚群，明确各亚群分子标志物与功能定位，填补膀胱癌 CAFs 亚群分类的研究空白，修正传统将 CAFs 视为单一群体的认知偏差； 2. 深入解析 CAFs 相关差异表达基因的调控网络及关键通路的作用，揭示CAFs 与肿瘤细胞、免疫细胞的交互规律，完善膀胱癌肿瘤微环境重塑理论； 3. 基于 CAFs 特征构建多终点预后评估指标，为后续研究提供标准与依据； 4. 为临床医生提供精准分层患者预后风险的工具，帮助调整随访策略，解决当前预后评估精准度不足的问题。  

Objectives of Study：

1. Systematically identify functional subpopulations of bladder cancer CAFs through multi-omics integrated analysis, clarify the molecular markers and functional localization of each subpopulation, fill the research gap in the classification of bladder cancer CAF subpopulations, and correct the traditional cognitive bias of viewing CAFs as a single population; 2. In-depth analysis of the regulatory networks of CAF-related differentially expressed genes and the roles of key pathways, revealing the interaction patterns between CAFs and tumor cells, as well as immune cells, thereby improving the theory of bladder cancer tumor microenvironment remodeling; 3. Construct multi-endpoint prognostic evaluation indicators based on CAF characteristics to provide standards and basis for subsequent research; 4. Provide clinicians with tools for precise stratification of patient prognostic risk, helping to adjust follow-up strategies and addressing the current issue of insufficient accuracy in prognostic evaluation.

数据采集 公共数据库数据：从 TCGA、GEO 等数据库下载膀胱癌患者的转录组数据、临床信息（含肿瘤分期、治疗方案、随访结局），通过标准化数据处理工具（如 R 语言 TCGAbiolinks 包）统一格式后提取有效信息。 本院样本数据：通过医院电子病历系统、病理科信息系统采集患者的临床病理资料（肿瘤分级、手术方式等）；通过实验检测（qPCR、免疫组化）获取 CAFs 相关基因表达数据，由实验人员双人录入 Excel 表格。 质量控制：公共数据需剔除重复、信息缺失的病例；本院临床数据由医护人员核对病历原件后录入，实验数据需记录操作时间、仪器参数，设重复实验验证可靠性。 数据管理 存储方式：所有数据匿名化处理（用编号替代患者姓名、身份证号），电子数据加密后存储于医院专用科研服务器，实验原始记录（纸质版）归档至科室资料室，由专人负责管理。 数据使用：仅授权研究团队成员访问数据，数据调用需记录使用目的与时间；研究结束后数据需至少保存 5 年，期间不得泄露给第三方或用于非本研究用途。 备份机制：电子数据定期（每周）进行本地 + 云端双重备份，防止数据丢失；纸质记录需防潮、防火，并存档副本。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：

Data Collection Public database data: Download transcriptome data and clinical information (including tumor stage, treatment plan, follow-up outcomes) of bladder cancer patients from databases such as TCGA and GEO, and extract valid information after unifying the format via standardized data processing tools (e.g., R package TCGAbiolinks). Hospital sample data: Collect clinicopathological data (tumor grade, surgical method, etc.) of patients through the hospital's electronic medical record system and pathology department information system; obtain CAFs-related gene expression data through experimental tests (qPCR, immunohistochemistry), which are double-entered into Excel spreadsheets by researchers. Quality control: Duplicate or incomplete cases are excluded from public data; hospital clinical data are entered after medical staff check the original medical records, and experimental data are recorded with operation time and instrument parameters, with repeated experiments to verify reliability. Data Management Storage method: All data are anonymized (patient names and ID numbers are replaced with codes); electronic data are encrypted and stored on the hospital's dedicated scientific research server; original experimental records (paper version) are filed in the department's data room and managed by a designated person. Data usage: Only authorized members of the research team can access the data; data access must be recorded with the purpose and time of use; data must be kept for at least 5 years after the study is completed, and shall not be disclosed to third parties or used for non-research purposes during this period. Backup mechanism: Electronic data are backed up locally and in the cloud regularly (weekly) to prevent data loss; paper records are protected from moisture and fire, and copies are filed.
