本研究旨在针对牙槽突裂修复中形态不匹配、骨整合困难与功能重建不足等关键瓶颈,提出并验证一套基于“3D打印个性化赋形技术协同BMAC负载β-TCP-CMC-Na支架”的精准再生策略。课题首先通过逆向工程与工艺优化,建立高精度、可复制的3D打印个性化模具与导板的标准化技术体系。在此基础上,开展一项单中心前瞻性三臂队列临床研究,对比评估该创新方案(A组)、去凝胶化对照方案(B组)与传统自体髂骨移植“金标准”(C组)的临床疗效,旨在系统验证CMC-Na凝胶的关键作用与3D打印个性化技术的综合优势,推动牙槽突裂治疗从“结构替代”向“精准重建”的范式转变,并为临床应用提供标准化解决方案。 |

Objectives of Study: |
This study aims to address the key bottlenecks in alveolar cleft (AC) repair—morphological mismatch, difficult osseointegration, and insufficient functional reconstruction—by proposing and validating a precision regeneration strategy based on "3D-printed personalized shaping technology combined with bone marrow aspirate concentrate (BMAC)-loaded beta-tricalcium phosphate (β-TCP)-carboxymethyl cellulose sodium (CMC-Na) scaffolds." The project first establishes a standardized technical system for high-precision, replicable 3D-printed personalized molds and guide plates through reverse engineering and process optimization. Building on this, a single-center, prospective three-arm cohort clinical study is conducted to comparatively evaluate the clinical efficacy of the innovative approach (Group A), a gel-free control approach (Group B), and the traditional autologous iliac bone graft "gold standard" (Group C). The study systematically verifies the critical role of CMC-Na gel and the comprehensive advantages of 3D-printed personalized technology, aiming to advance the paradigm shift in alveolar cleft treatment from "structural substitution" to "precision reconstruction" and provide a standardized solution for clinical application. |

1.经影像学确诊单侧牙槽突裂; 2.未接受过牙槽突裂修复治疗; 3.无骨代谢异常; 4.术区无感染; 5.研究参与者和/或其监护人同意参加本试验,并签署知情同意书; 6.未同时参与其他临床试验。 |

Inclusion criteria |
1. Unilateral alveolar cleft confirmed by imaging examination. 2. No prior surgical repair for alveolar cleft. 3. No evidence of bone metabolic disorders. 4. No signs of infection in the surgical area. 5. The participant and/or their legal guardian agrees to participate in this trial and provides signedinformed consent. 6. Not currently participating in any other clinical trial. |

排除标准: |
1.既往牙槽突裂修复史; 2.上颌骨手术史; 3.存在全身感染或手术局部感染; 4.具有其他可能影响骨愈合的骨代谢性疾病的病史或相关检查异常(如骨质疏松、骨软化症等); 5.术前检查及评估全身情况无法耐受全麻手术以及其他原因不适宜手术者; 6.无法配合完成术前评估及试验期内相关检查者; 7.正在参与其他临床试验,且未达到终点者。 |

Exclusion criteria: |
1. History of previous surgical repair for alveolar cleft. 2. History of previous maxillary surgery. 3. Presence of systemic infection or local infection at the surgical site. 4. History or related examination abnormalities of other bone metabolic diseases (e.g.,osteoporosis, osteomalacia)that may affect bone healing. 5. Preoperative evaluation indicating intolerance to general anesthesia or any other conditionrendering the patient unsuitable for surgery. 6. Inability to cooperate with preoperative assessments or required examinations during the trialperiod. 7. Currently participating in another clinical trial and has not yet reached the study endpoint. |
