研究目的：

1. 全面挖掘和筛选与肺部肿瘤发生发展高度相关的重要眼影像学标志物，阐述“眼-肺轴”的表型关联性，为疾病的无创筛查及早期预警提供客观、便捷的新型影像学生物标志物。 2. 探索眼影像学标志物与肺部肿瘤的关联机制，明确二者共享遗传风险位点、遗传相关性，同时锚定炎症因子、血浆蛋白质、代谢产物水平作为眼影像学标志物与肺部肿瘤的中介因素，运用孟德尔随机化分析探索因果关系及其潜在的中介效应，阐述“眼-肺轴”的基因关联性及蛋白、代谢关联性，提高以眼观肺部肿瘤的可解释性。 3. 在现有疾病风险预测模型基础上，整合筛选出的关键眼影像学标志物，构建基于眼影像组学的肺部肿瘤筛查模型、诊断判别模型和预后预测模型：筛查模型旨在用于肺部肿瘤高危人群的初筛，以优化LDCT筛查流程；诊断判别模型旨在辅助就诊者的肺部肿瘤风险分层；预后模型旨在用于患病人群的治疗相关并发症和远期预后预测，助力疾病风险分层及预后评估的无创化、精准化、高效化。  

Objectives of Study：

1. Identify key ophthalmic imaging biomarkers closely associated with the development and progression of pulmonary tumors, elucidate the phenotypic correlation of the "eye-lung axis," and provide objective and convenient biomarkers for non-invasive screening and early warning. 2. Investigate the mechanisms linking ophthalmic imaging biomarkers with pulmonary tumors, including shared genetic risk loci and genetic correlations. Inflammatory factors, plasma proteins, and metabolites will be evaluated as potential mediators, and Mendelian randomization will be used to explore causal relationships and mediating effects, thereby clarifying the genetic, proteomic, and metabolic basis of the "eye-lung axis" and improving the interpretability of ocular-based pulmonary tumor prediction. 3. Develop and validate ophthalmic imaging radiomics-based screening, diagnostic, and prognostic models for pulmonary tumors by integrating the selected key biomarkers. These models will support initial screening in high-risk populations, assist risk stratification in clinical settings, and predict treatment-related complications and long-term outcomes, enabling non-invasive, precise, and efficient risk assessment and prognostic evaluation.

纳入标准：

病例组： 1. 由肺外科、肺肿瘤科综合临床症状、肺部影像学检查、组织活检等检查明确诊断为肺癌（ICD-10编码C34）的患者。 2. 年龄>=18周岁，自愿参加并签署知情同意书。 健康对照组： 1.体检中心招募的同样年龄、性别构成且无眼病和肺部肿瘤的受试者，允许合并常见慢性疾病。

Inclusion criteria

Case Group: 1. Patients who have been definitively diagnosed with lung cancer (ICD-10 code C34) based on a comprehensive evaluation of clinical symptoms by thoracic surgery and thoracic oncology specialists, lung imaging studies, and tissue biopsy. 2. Aged 18 years or older, voluntarily participating and having signed an informed consent form. Healthy Control Group: 1. Subjects recruited from the health examination center with the same age and gender composition, without eye diseases or lung tumors, allowing for the presence of common chronic diseases.

排除标准：

1. 合并有严重眼部疾病或接受过眼内手术。 2. 因全身疾病或个人原因无法配合或完成全部的眼科检查。 3. 具有任何可能影响方案依从性的疾病史（如严重精神障碍、认知功能障碍、药物滥用或成瘾等）。

Exclusion criteria：

1. Patients with significant ocular comorbidities or a history of intraocular surgery. 2. Inability to complete required ophthalmic examinations due to systemic conditions or personal reasons. 3. Medical history that may compromise protocol compliance, including: severe psychiatric disorders,cognitive impairment, substance abuse or addiction.
