研究目的：

（1）明确COPD患者BALF中LC3、Beclin-1、PINK1/Parkin的表达特征，并与健康对照组比较。 （2）分析自噬相关蛋白与疾病严重程度（GOLD分级、急性加重频率、CAT评分）的相关性，评估其作为生物标志物的潜力。 （3）探索自噬-炎症交互作用，如LC3与IL-6、TNF-α的关系，揭示自噬调控COPD炎症的机制。 （4）结合PINK1/Parkin表达，评估线粒体自噬在COPD肺损伤中的作用，为靶向干预提供依据。  

Objectives of Study：

(1) Determine the expression characteristics of LC3, Beclin-1, and PINK1/Parkin in BALF of COPD patients and compare them with those of a healthy control group. (2) Analyze the correlation between autophagy-related proteins and disease severity (GOLD grading, frequency of acute exacerbations, CAT score), and evaluate their potential as biomarkers. (3) Explore the autophagy-inflammatory interaction, such as the relationship between LC3 and IL-6, TNF-α, to reveal the mechanism by which autophagy regulates inflammation in COPD. (4) Combine PINK1/Parkin expression to evaluate the role of mitophagy in COPD-related lung injury, providing a basis for targeted intervention.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.年龄＞=40岁，且存在慢性咳嗽、咳痰、呼吸困难等典型呼吸道症状；
2.依据2024年慢性阻塞性肺疾病全球倡议（GOLD）诊断标准，使用支气管舒张剂后第1秒用力呼气容积（FEV1）/用力肺活量（FVC）<70%；
3.自愿参与本研究，签署知情同意书，并能够配合完成肺泡灌洗、肺功能检查及相关随访；

Inclusion criteria

1.Age ＞= 40 years, with typical respiratory symptoms such as chronic cough, sputum production, and dyspnea;
2.According to the 2024 Global Initiative for Chronic Obstructive Lung Disease (GOLD) diagnostic criteria, post-bronchodilator FEV1/FVC < 70%;
3.Willing to participate in this study, having signed an informed consent form, and able to cooperate in completing bronchoalveolar lavage, pulmonary function tests, and related follow-ups;

排除标准：

1.合并其他影响肺部功能的疾病，如间质性肺疾病、支气管哮喘、肺结核、肺部恶性肿瘤等；
2.近3个月内使用过免疫抑制剂、全身性糖皮质激素（剂量超过泼尼松10mg/d）；
3.存在严重的心、肝、肾等重要脏器功能衰竭；
4.妊娠或哺乳期女性；
5.有精神疾病或认知障碍，无法配合研究流程；
6.近期（≤1个月）发生过急性加重或接受过机械通气治疗；

Exclusion criteria：

1.Combined with other diseases affecting lung function, such as interstitial lung disease, bronchial asthma, tuberculosis, malignant lung tumors, etc; 2.Use of immunosuppressants or systemic glucocorticoids (at a dose exceeding prednisone 10mg/day) within the past 3 months; 3.Presence of severe functional failure in vital organs such as the heart, liver, or kidneys; 4.Pregnant or breastfeeding women; 5.Presence of mental illness or cognitive impairment, unable to cooperate with the research process; 6.Recent (<=1 month) occurrence of acute exacerbation or receipt of mechanical ventilation treatment.
