研究目的：

本研究旨在建立一套可重复的、人源的神经化皮肤类器官模型，该模型由原代人角质形成细胞、原代人真皮成纤维细胞和诱导多能干细胞（iPSC）-来源的痛觉神经元（nociceptor）构成，验证模型在结构学（如表皮分层、真皮ECM与感觉神经纤维伸入）与功能学（如TRPV1介导的Ca2+响应、神经肽释放与药物反应）方面的可用性，并评估其在疼痛药物与抗炎药筛选、神经—免疫互作研究等下游应用的适用性。  

Objectives of Study：

The aim of this study was to establish a reproducible, human-derived neuralized skin organoid model, the model consisted of primary human keratinocyte, primary human dermal fibroblasts, and induced pluripotent stem cell (iPSC)-derived pain-sensing neurons (nociceptors) , which were cultured in vitro, to validate the availability of the model in terms of structural (such as epidermal stratification, dermal ECM with sensory nerve fiber extension) and functional (such as TRPV1-mediated Ca2 + response, neuropeptide release with drug response) aspects, and to provide a theoretical basis for the development of the model, and to evaluate its suitability for downstream applications such as the screening of pain drugs and anti-inflammatory drugs, and the study of neuro-immune interactions.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.年龄18～55岁，拟行择期手术（如整形/美容手术等）；
2.意识清楚，具备完全民事行为能力，能够理解研究内容并自愿签署书面知情同意书。
3.术前根据医院常规检查，无活动性全身严重感染。
4.术区皮肤外观基本正常，无明显急性化脓性感染、严重湿疹、大片皮肤溃疡等。
5.同意其手术废弃皮肤组织用于本研究，并同意在去标识化后进行体外实验研究。

Inclusion criteria

1. 18-55 years of age, scheduled for elective surgery (e.g. plastic/cosmetic surgery); 2. Conscious and fully capable of civil conduct, able to understand the research content and voluntarily sign a written informed consent form. 3. Based on routine hospital examinations prior to surgery, there is no active systemic severe infection. 4. The skin appearance in the surgical area is generally normal, with no signs of acute suppurative infection, severe eczema, or extensive skin ulcers. 5. Consent to the use of discarded skin tissue from their surgery for this study, and consent to its use in in vitro experimental research after de-identification.

排除标准：

1.无法充分理解研究信息，或存在精神、认知障碍，无法表达真实意愿者；
2.术区存在明显急性化脓性感染、严重皮肤炎症、恶性肿瘤或疑似恶性病变；
3.既往有严重出血性疾病或凝血功能明显异常，需改变手术方式或切除范围者；
4.术前评估存在严重全身性疾病（如失代偿期心、肝、肾功能不全）导致手术风险明显高于常人者；
5.已知合并HIV感染或其他高度传染性疾病，且临床不建议将其组织进入一般实验流程者；
6.不愿意参加本研究或拒绝签署知情同意书者；
7.研究者认为不适合参与本研究的其他情况；

Exclusion criteria：

1.Individuals who are unable to fully comprehend research information, or who have mental or cognitive impairments that prevent them from expressing their true wishes.
2.The surgical site exhibits significant acute suppurative infection, severe skin inflammation, malignant tumors, or suspected malignant lesions.
3.Patients with a history of severe hemorrhagic disorders or significant coagulation abnormalities requiring modification of surgical approach or resection extent.
4.Patients with severe systemic diseases (such as decompensated cardiac, hepatic, or renal insufficiency) identified during preoperative assessment, resulting in significantly higher surgical risks than the general population.
5.Individuals known to have HIV infection or other highly infectious diseases, whose tissue is not clinically recommended for inclusion in standard laboratory procedures.
6.Individuals unwilling to participate in this study or who refuse to sign the informed consent form;
7.Other circumstances deemed unsuitable for participation in this study;
