本项目旨在系统解析骨科手术老年患者中肌少症的分子基础。具体目标包括:全景式描绘肌少症多组学分子图谱;鉴定与肌少症密切相关的差异分子;解析核心病理通路与关键调控节点;构建肌少症的整合性分子调控网络,以期为新型生物标志物和药物靶点的发现提供依据。  

Objectives of Study:

This project aims to systematically elucidate the molecular basis of sarcopenia in elderly orthopaedic surgery patients. Specific objectives include: - Constructing a comprehensive multi-omics molecular landscape of sarcopenia; - Identifying differentially expressed molecules closely associated with sarcopenia; - Analysing core pathological pathways and key regulatory nodes; - Establishing an integrated molecular regulatory network for sarcopenia, thereby providing a foundation for the discovery of novel biomarkers and therapeutic targets.Translated with DeepL.com (free version)

Description for medicine or protocol of treatment in detail:

 

纳入标准:

1.年龄 ≥ 60周岁,性别不限。
2.计划在未来3-12个月内接受择期下肢骨科手术的患者。
3.意识清楚,能够理解研究内容并独立签署知情同意书,或其法定代理人同意。
4.能够配合完成肌力、躯体功能等相关检查。
5.BMI范围:18.5 kg/m2 ≤ BMI < 35.0 kg/m2,以排除营养不良或重度肥胖对肌肉代谢的极端影响。

Inclusion criteria

1. Age >= 60 years, no gender restrictions. 2. Patients scheduled to undergo elective lower limb orthopaedic surgery within the next 3–12 months. 3. Conscious and able to comprehend the study content and independently sign the informed consent form, or have consent from their legal representative. 4. Able to cooperate with relevant examinations including muscle strength and physical function assessments. 5. Body Mass Index (BMI) range: 18.5 kg/m2 <= BMI < 35.0 kg/m2, to exclude extreme effects of malnutrition or severe obesity on muscle metabolism.

排除标准:

1.患有严重影响肌肉代谢或功能的疾病:如未控制的甲状腺功能亢进/减退、库欣综合征、严重慢性肾脏病(eGFR < 30 mL/min/1.73m2)、严重肝功能不全、失代偿性心力衰竭。
2.神经肌肉疾病:如帕金森病、肌萎缩侧索硬化、重症肌无力、遗传性肌病等。
3.系统性炎症性疾病:如类风湿性关节炎、系统性红斑狼疮等。
4.长期(>3个月)口服或注射糖皮质激素或其他已知会影响肌肉质量的药;
5.营养不良,恶液质状态。
6.骨科急诊手术患者。
7.有酒精或药物滥用史。
8.无法完成必要的基线评估或拒绝提供肌肉样本。

Exclusion criteria:

1. Conditions severely affecting muscle physiology or function: such as uncontrolled hyperthyroidism/hypothyroidism, Cushing's syndrome, severe chronic kidney disease (eGFR < 30 mL/min/1.73m2), severe hepatic impairment, decompensated heart failure. 2. Neuromuscular disorders: e.g., Parkinson's disease, amyotrophic lateral sclerosis, myasthenia gravis, hereditary myopathies. 3. Systemic inflammatory diseases: e.g., rheumatoid arthritis, systemic lupus erythematosus. 4. Long-term (>3 months) oral or injectable glucocorticoid therapy or other medications known to affect muscle mass. 5. Malnutrition, cachectic state. 6. Patients undergoing orthopaedic emergency surgery. 7. History of alcohol or substance abuse. 8. Inability to complete necessary baseline assessments or refusal to provide muscle samples.
