研究目的：

利用非代谢组学分析，了解 OA 患者血清及软骨细胞中代谢物的变化，进行KEGG富集分析，WB、软骨组织 IHC 染色对比磨损侧与非磨损侧软骨组织差异。  

Objectives of Study：

Untargeted metabolomics was employed to profile metabolic alterations in serum and chondrocytes of OA patients. KEGG enrichment analysis was performed, followed by Western blotting (WB) and immunohistochemical (IHC) staining of articular cartilage tissues to contrast differences between weight-bearing (wear-side) and non-weight-bearing areas.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.在莆田学院附属医院行单侧全膝关节置换术的终末期膝骨关节炎（Kellgren-Lawrence IV级）患者； 2.年龄18-80岁； 3.术前6月未使用免疫抑制剂或关节腔注射； 4.签署知情同意书。

Inclusion criteria

1. Patients with end-stage knee osteoarthritis (Kellgren-Lawrence grade IV) who underwent unilateral total knee arthroplasty at Putian University Affiliated Hospital; 2. Age range: 18-80 years old; 3. No use of immunosuppressants or intra-articular injections for 6 months before the surgery; 4. Signed the informed consent form..

排除标准：

1.合并类风湿关节炎、痛风性关节炎、创伤性关节炎或感染性关节炎； 2.严重肝肾功能不全（Child-Pugh C级/eGFR<30）； 3.恶性肿瘤病史； 4.术中确诊软骨钙化或广泛骨坏死； 5.拒绝提供组织样本。

Exclusion criteria：

1. Include rheumatoid arthritis, gouty arthritis, traumatic arthritis or infectious arthritis; 2. Severe liver and kidney dysfunction (Child-Pugh C grade / eGFR < 30); 3. History of malignant tumor; 4. Confirmed intraoperative cartilage calcification or extensive bone necrosis; 5. Refusal to provide tissue samples.
