研究目的:

(1)妊娠期维生素 A、D、E营养状态可能与葡萄糖代谢相关; (2)妊娠期糖尿病患者可能存在氧化应激失衡,造成胰岛素β细胞功能障碍,使血糖调节受到影响,其中8-羟基脱氧鸟苷酸(8-OHdG)是DNA氧化损伤的特异及主要的产物,目前已成为DNA氧化损失中最常用的生物标记物; (3)维生素 A、D、E是具有抗氧化功能的脂溶性维生素,主要参与了机体抗氧化过程,如缺乏可导致体内发生氧化应激反应。结合以上三点,本课题组提出维生素 A、D、E 的缺乏导致体内8-0HdG升高,最终发生GDM的假设,采用前瞻性病例对照的研究方法,利用生物素双抗体夹心酶联免疫吸附法(ELISA)测定血液8-0HdG水平,进行相关性研究Pearson相关系数分析,通过病例之间的数据比较,验证是否妊娠期糖尿病患者维生素A、D、E缺乏,导致机体发生氧化应激反应,使的体内8-0HdG产生增加。为GDM的预测及早期干预提供新的理论依据。  

Objectives of Study:

(1) The nutritional status of vitamins A, D, and E during pregnancy may be associated with glucose metabolism; (2) Patients with gestational diabetes mellitus may have an imbalance of oxidative stress, which leads to pancreatic β-cell dysfunction and impairs blood glucose regulation. Among relevant indicators, 8-hydroxy-2'-deoxyguanosine (8-OHdG) is a specific and major product of DNA oxidative damage, and has now become the most commonly used biomarker for DNA oxidative damage; (3) Vitamins A, D, and E are fat-soluble vitamins with antioxidant functions, which are mainly involved in the body's antioxidant process. Their deficiency can induce oxidative stress responses in the body. Based on the above three points, our research team puts forward the hypothesis that the deficiency of vitamins A, D, and E leads to an increase in the level of 8-OHdG in the body, ultimately resulting in GDM. A prospective case-control study was adopted, and the enzyme-linked immunosorbent assay (ELISA) with a biotin double-antibody sandwich method was used to determine the serum 8-OHdG level. we verified whether the deficiency of vitamins A, D, and E in patients with gestational diabetes mellitus induces oxidative stress responses in the body, thereby increasing the production of 8-OHdG. This study is expected to provide new theoretical basis for the prediction and early intervention of GDM.

1.孕期OGTT试验阳性; 2.宫内单活胎。

Inclusion criteria

1. Positive result on the oral glucose tolerance test (OGTT) during pregnancy; 2. Singleton live fetus in utero.

排除标准:

1.患有其他基础疾病未经治愈患者; 2.妊娠期糖尿病需药物治疗或孕前糖尿病患者。

Exclusion criteria:

1. Patients with other underlying diseases that are not cured; 2. Patients with gestational diabetes requiring medication or pre-pregnancy diabetes.
