本研究通过采用无标记定量蛋白质组学技术对TBM与非TBM患者脑脊液蛋白进行比较分析,筛选出差异蛋白,进一步通过Western blot技术进行候选蛋白验证,筛选出的差异蛋白联合前期研究基础筛选出的13个差异蛋白进行Elisa验证,结合患者临床资料,利用非线性的支持向量机(Support Vector Machine, SVM)分类器分析患者临床资料及筛选出的蛋白,建立诊断模型,评估模型诊断价值,发现相对简便、高效的诊断模型。 |

Objectives of Study: |
In this study, unmarked quantitative proteomics technology will be employed to compare the protein levels in the cerebrospinal fluid of TBM patients and non-TBM patients. The differentially expressed proteins are identified. Further, candidate proteins will be verified by Western blot technique. The identified differentially expressed proteins are combined with the 13 differentially expressed proteins screened out from the previous study for Elisa verification. Combined with the clinical data of the patients, a non-linear support vector machine (SVM) classifier is used to analyze the clinical data and the screened proteins, and a diagnostic model will be established. The diagnostic value of the model will be evaluated. A relatively simple and efficient diagnostic model will be discovered. |

1.北京胸科医院急诊及住院疑似TBM患者。 2.TBM患者抗结核治疗小于2周,其他中枢神经系统感染患者治疗小于1周。 3.无腰椎穿刺禁忌症 4.患者同意入组,并签署知情同意书。 |

Inclusion criteria |
1. Emergency and inpatient patients with suspected TBM at Beijing Chest Hospital. 2.Patients with TBM who have received anti-tuberculosis treatment for less than 2 weeks, and patients with other central nervous system infections who have received treatment for less than 1 week. 3. There are no contraindications for lumbar puncture. 4. The patients have agreed to participate in the study and have signed the informed consent form. |

排除标准: |
1.诊断为可能结核性脑膜炎患者。 2.不能明确诊断患者。 3.脑外伤或颅内有创检查导致颅内感染者。 4.长期使用糖皮质激素、非甾体抗炎药,或免疫抑制剂。 |

Exclusion criteria: |
1. Patients diagnosed with possible tuberculous meningitis. 2. Patients whose diagnosis is unclear. 3. Infections in the brain caused by brain trauma or invasive intracranial examinations. 4. Long-term use of glucocorticoids, non-steroidal anti-inflammatory drugs, or immunosuppressants. |
