通过收集患儿的临床表型资料,并结合家系成员的外周血样本进行 外显子组测序与转录组测序,验证该变异对 CREBBP 剪接的实际影响,以明确其是否导致功能丧失并判断其致病性。  

Objectives of Study:

By collecting clinical phenotype data from affected children and combining it with peripheral blood samples from family members for exome sequencing and transcriptome sequencing, we validated the actual impact of this variant on CREBBP splicing to determine whether it causes loss of function and assess its pathogenicity.

The study includes sensitive individual-level genomic and clinical data. To protect participant privacy, raw data will not be publicly deposited. De-identified summary results will be published. Raw data may be shared only with authorized research collaborators under data use agreements and subject to ethics review.

数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC:

研究数据包括参与者基本信息、临床表型、辅助检查结果及分子检测数据(外显子测序和 RNA-seq 结果)。所有数据按统一标准化表格采集,由受过培训的研究人员录入电子数据库,设置逻辑校验以保证准确性。数据在采集后由专人定期检查,发现异常及时更正。 所有数据进行 去标识化处理,仅保留研究编号。数据存储于安全的服务器和受限权限的实验室电脑中,并定期备份。数据库访问仅限授权研究团队成员,并通过权限控制和密码保护确保数据安全与保密。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture:

Study data include participants’ basic demographic information, clinical phenotypes, auxiliary examination results, and molecular test data (exome sequencing and RNA-seq). All data are collected using standardized forms and entered into an electronic database by trained research staff, with built-in checks to ensure data accuracy. Data quality is regularly reviewed and corrected as needed.All data are de-identified and stored on a secure server and controlled-access laboratory computers with routine backup. Access to the database is limited to authorized study team members through role-based permissions and password protection to ensure data security and confidentiality.
