研究目的：

对比正常对照组与睡眠障碍组间肠道、鼻腔微生物以及代谢物的差异，以期发现新的睡眠障碍生物标志物。  

Objectives of Study：

Compare the differences in gut and nasal microbiota, as well as metabolites, between the normal control group and the sleep disorder group in order to discover new sleep disorder biomarkers.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1. 健康对照组：(1) 年龄 18-65 周岁；(2) 匹兹堡睡眠质量指数（PSQI）总分<= 5；(3) 阿森斯失眠量表总分< 4；(4) 来源于睡眠障碍组中某位索引患者的一级家属（父母、子女、兄弟姐妹）；(5) 情绪健康：汉密尔顿抑郁量表（HAMD-17）得分< 7；(6) 汉密尔顿焦虑量表（HAMA）得分< 7；(7) 无精神病史：无任何精神障碍当前或既往史； 2. 睡眠障碍组：(1) 年龄 18-65 周岁；(2) 匹兹堡睡眠质量指数（PSQI）总分> 7；(3) 阿森斯失眠量表总分> 4

Inclusion criteria

1. Healthy control group: (1) Aged 18 to 65 years old; (2) The total score of the Pittsburgh Sleep Quality Index (PSQI) is <= 5; (3) The total score of the Parsons Insomnia Scale is less than 4; (4) From the first-level family members (parents, children, siblings) of a certain index patient in the sleep disorder group; (5) Emotional health: Hamilton Depression Scale (HAMD-17) score < 7; (6) Hamilton Anxiety Scale (HAMA) score < 7; (7) No history of mental illness: No current or past history of any mental disorders; 2. Sleep disorder group: (1) Aged 18 to 65 years old; (2) The total score of the Pittsburgh Sleep Quality Index (PSQI) is > 7; (3) The total score of the Parsons Insomnia Scale is greater than 4

排除标准：

1. 鼻腔局部因素：慢性鼻炎、鼻窦炎、鼻息肉、过敏性鼻炎急性发作期、近期（4周内）鼻腔手术或外伤史。 2. 胃肠道局部因素：炎症性肠病、肠易激综合征、慢性便秘或腹泻（罗马IV标准）、胃肠道恶性肿瘤史、近期（3个月内）重大胃肠道手术史。 3. 重大精神神经疾病：精神分裂症、双相情感障碍、创伤后应激障碍、帕金森病、癫痫、痴呆等。 4. 干扰性用药史：(1) 近4周内使用过抗生素、益生菌、益生元、合生元。(2) 近3个月内规律使用影响胃肠道功能的药物（如质子泵抑制剂、促胃肠动力药）。(3) 近3个月内使用过免疫抑制剂或皮质类固醇。(4) 系统性疾病：自身免疫性疾病、未控制的高血压、糖尿病、甲状腺功能异常、恶性肿瘤、严重心肝肾衰竭。(5) 近期感染与特殊生理状态：近4周内有急性感染或发热史；怀孕或哺乳期女性。(6) 生活与饮食习惯：长期轮班、夜班工作者；有酗酒史或药物滥用史；近期有重大饮食结构改变。

Exclusion criteria：

1. Local factors of nasal cavity: chronic rhinitis, sinusitis, nasal polyps, acute exacerbation of allergic rhinitis, recent (within 4 weeks) history of nasal surgery or trauma. 2. Local gastrointestinal factors: inflammatory bowel disease, irritable bowel syndrome, chronic constipation or diarrhea (Rome IV criteria), history of gastrointestinal malignancies, recent (within 3 months) major gastrointestinal surgery history. 3. Major mental and neurological disorders: schizophrenia, bipolar disorder, post-traumatic stress disorder, Parkinson's disease, epilepsy, dementia, etc. 4. History of interfering medication: (1) Used antibiotics, probiotics, prebiotics, and synbiotics in the past 4 weeks. (2) Regularly using drugs that affect gastrointestinal function (such as proton pump inhibitors and prokinetic drugs) within the past 3 months. (3) Have used immunosuppressants or corticosteroids in the past 3 months. (4) Systemic diseases: autoimmune diseases, uncontrolled hypertension, diabetes, thyroid dysfunction, malignant tumors, severe heart, liver and kidney failure. (5) Recent infection and special physiological status: History of acute infection or fever in the past 4 weeks; Pregnant or lactating women. (6) Lifestyle and dietary habits: long-term shift and night shift workers; Have a history of alcoholism or drug abuse; There have been significant changes in dietary structure recently.
