研究目的：

本研究拟基于肝癌患者手术切除后剩余的癌旁肝组织样本，系统评估KLB信号通路在癌旁组织中的表达特征，并分析其与细胞周期相关分子（如Cyclin D1）及免疫抑制/免疫调控分子（如CD47等）的相关性，旨在阐明KLB参与肝癌免疫微环境调控的潜在机制，为后续靶向KLB的治疗策略提供机制依据。  

Objectives of Study：

To investigate the expression profile of the KLB signaling pathway in peritumoral liver tissues from hepatocellular carcinoma patients and to analyze its associations with cell-cycle regulators (e.g., Cyclin D1) and immune-modulatory molecules (e.g., CD47). This study aims to elucidate the potential role of KLB in shaping the tumor immune microenvironment and to provide mechanistic evidence for future KLB-targeted therapeutic strategies.

纳入标准：

1.年龄 ≥18 岁，男女不限； 2.拟在重庆大学附属三峡医院接受肝癌根治性切除术或经皮肝穿刺活检，手术方案中可获得足量癌旁组织； 3. 影像学或病理学提示为肝细胞癌（HCC），具备采集癌旁组织的条件； 4.志愿者自愿配合该研究并签署知情同意书； 5.临床状态稳定，符合组织采集要求，无重大术前禁忌

Inclusion criteria

1. Age >=18 years, regardless of gender; 2. Planning to undergo radical resection or percutaneous liver biopsy for liver cancer at the Third Gorges Hospital affiliated with Chongqing University, with sufficient adjacent non-cancerous tissue available according to the surgical plan; 3. Imaging or pathology indicates hepatocellular carcinoma (HCC), and conditions for collecting adjacent non-cancerous tissue are met; 4. The volunteer voluntarily agrees to participate in this study and signs the informed consent form; 5. Clinical condition is stable, meeting tissue collection requirements, with no major preoperative contraindications.

排除标准：

1.病理无法确认癌旁组织，或组织样本质量不足以满足实验需求； 2.合并严重基础疾病（如系统性免疫病、活动性感染等），影响组织免疫背景判断； 3.曾接受肝移植，或正在接受系统性免疫抑制治疗者； 4.妊娠或哺乳期女性； 5.拒绝签署知情同意书，或签署后中途撤回授权者

Exclusion criteria：

1. Pathology cannot confirm cancer-adjacent tissue, or the tissue sample quality is insufficient to meet experimental requirements; 2. Combined severe underlying diseases (such as systemic immune diseases, active infections, etc.) that affect the assessment of tissue immune background; 3. Have undergone liver transplantation, or are currently receiving systemic immunosuppressive therapy; 4. Pregnant or breastfeeding women; 5. Refuse to sign the informed consent, or withdraw authorization after signing.

临床与病理信息由研究人员通过病历系统提取并编码匿名化；组织样本检测数据（IHC/WB/qPCR/RNA-seq）由实验室按统一SOP获取。所有数据采用双人核对录入，存储于医院科研平台的加密电脑/服务器，限定授权访问，定期备份。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：

Clinical/pathological data will be extracted from the hospital EMR and coded. Molecular data (IHC/WB/qPCR/RNA-seq) will be generated following standardized SOPs. Data will be double-checked, stored on password-protected hospital research servers with restricted access, and regularly backed up.
