研究目的：

1. 确证维生素D3联合DHA对MCI老年人总体认知功能（以韦氏成人智力量表总智商为主要终点）的改善效果； 2. 探究联合干预与单一干预效果的差异； 3. 阐明干预措施对外周血NLRP3炎症小体通路及相关炎症标志物的影响，从而在人群层面为其神经免疫机制提供证据线索。  

Objectives of Study：

1. Confirm the improvement effect of vitamin D3 combined with DHA on the overall cognitive function of elderly people with MCI (with the total IQ of the Wechsler Adult Intelligence Scale as the main endpoint); 2. Explore the differences in the effectiveness of joint intervention and single intervention; 3. Clarify the impact of intervention measures on the NLRP3 inflammasome pathway and related inflammatory markers in peripheral blood, in order to provide evidence clues for its neuroimmune mechanisms at the population level.

纳入标准：

1.参与者主观认为或由家庭成员/医生客观证实存在记忆减退； 2.记忆减退病程持续至少三个月； 3.简易精神状态检查 (MMSE) 分数文盲组≥17分、小学组≥20分、中学及以上组≥24分； 4.根据日常生活活动量表测量，日常生活活动没有或受到轻微损害，ADL得分≤18分； 5.不符合痴呆、AD、精神疾病、脑损伤或可能导致认知缺陷和任何活动性神经精神疾病的标准。 痴呆诊断标准按照美国精神疾病学会的DSM-IV标准和美国神经学、语言障碍和卒中-老年期痴呆和相关疾病学会（NINCDS-ADRDA）标准，并结合病史等资料进行痴呆诊断。采用HIS鉴别AD及血管性痴呆。可疑MCI及老年性痴呆患者由神经科医师进一步作出诊断。

Inclusion criteria

1. Participants subjectively believe or are objectively confirmed by family members/doctors to have memory impairment; 2. Memory impairment lasts for at least three months; 3. The Mini Mental State Examination (MMSE) score is>= 17 for the illiterate group, >= 20 for the primary school group, and >=24 for the middle school and above group; According to the Activities of Daily Living Scale, if there is no or slight impairment in daily living activities, the ADL score is <= 18 points; 5. Not suitable for dementia AD The criteria for mental illness, brain injury, or any active neuropsychiatric disorder that may lead to cognitive impairment. The diagnostic criteria for dementia are based on the DSM-IV criteria of the American Psychiatric Association and the National Institute of Neurology, Language Disorders, and Stroke Alzheimer's and Related Disorders (NINCDS-ADRDA) criteria, combined with medical history and other data for dementia diagnosis. Using HIS to differentiate between AD and vascular dementia. Suspected MCI and senile dementia patients will be further diagnosed by neurologists.

排除标准：

1. 临床诊断为 AD、PD 或其他痴呆症； 2. 存在会影响认知功能的严重器官功能障碍、恶性肿瘤或 其他重大疾病； 3. 在招募前 3 个月内服用过会干扰维生素D3 或 DHA 代谢的营养补充剂； 4. 有禁止服用维生素D3 或 DHA 的医疗状况。

Exclusion criteria：

1. Clinical diagnosis of AD, PD or other dementia; 2. Having serious organ dysfunction, malignant tumors, or other major diseases that can affect cognitive function; 3. Have taken nutritional supplements that interfere with vitamin D3 or DHA metabolism within the past 3 months prior to recruitment. 4. Medical conditions that prohibit the use of vitamin D3 or DHA.

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：

一、数据采集：以面对面访问的形式进行问卷调查对基本信息及采集数据进行收集，避免无效问卷。 量表：使用《韦氏成人智力量表中国修订版》标准化量表进行认知评估，确保数据有效。 流程：基于课题组前期的队列研究社区调查基础，招募符合标准并自愿参与研究的研究对象。 二、数据管理录入清洗：采用双录入模式从问卷直接进行原始数据录入，用 SPSS26.0 剔除填写异常、漏填的无效数据，统一编码转换为分析用数据。 存储备份：原始数据和清洗后数据均加密存储，同步备份至加密电脑，定期备份防丢失。 三、数据分析用 SPSS26.0 进行统计分析，先描述研究对象基本特征和量表指标，再根据研究需求做组间比较、指标关联或影响因素分析。 四、数据安全数据匿名化处理，删除个人识别信息，仅用于本研究。 遵循伦理要求，数据不外泄、不另作他用。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：

1. Data collection: Conduct a questionnaire survey in the form of face-to-face interviews to collect basic information and collected data, in order to avoid invalid questionnaires. Scale: Use the standardized Wechsler Adult Intelligence Scale (Chinese Revised Edition) for cognitive assessment to ensure data validity. Process: Based on the preliminary queue research community survey conducted by the research group, recruit research subjects who meet the criteria and voluntarily participate in the study. 2. Data management input cleaning: Adopting a dual input mode to directly input raw data from the questionnaire, using SPSS 26.0 to remove invalid data with abnormal or missing filling, and uniformly coding and converting it into data for analysis. Storage backup: Both raw data and cleaned data are encrypted and stored, synchronously backed up to encrypted computers, and regularly backed up to prevent loss. 3 Data analysis was conducted using SPSS 26.0 for statistical analysis. First, the basic characteristics and scale indicators of the research subjects were described, and then inter group comparisons, indicator associations, or analysis of influencing factors were conducted according to research needs. 4 Data security data anonymization processing, deletion of personal identification information, only used for this study. Adhere to ethical requirements, data should not be leaked or used for other purposes.
