研究目的:

本研究旨在通过多组学测序技术,深入探讨肺毛霉菌病的免疫机制及其发病过程。我们计划收集50例肺泡灌洗液、外周血样本及粪便,其中肺毛霉菌病组30例,单纯肺小结节组(对照组)20例。研究目标是通过单细胞测序、微生物组分析、代谢组学及空间转录组等多组学手段,揭示肺毛霉菌病中免疫细胞的种类、功能及其亚群特征,分析免疫细胞在该病理过程中所起的作用及其调控机制。通过这些分析,我们希望能够更加清晰地阐明免疫细胞如何参与肺毛霉菌病的免疫应答,为疾病的早期诊断、精准治疗及免疫调控策略的开发提供理论依据和数据支持。  

Objectives of Study:

This study aims to explore the immune mechanisms and pathogenesis of pulmonary aspergillosis using multi-omics sequencing technologies. We plan to collect alveolar lavage fluid, peripheral blood samples, and feces from 50 cases, with 30 cases in the pulmonary aspergillosis group and 20 cases in the simple pulmonary nodule group (control group). The goal of the study is to reveal the types, functions, and subpopulation characteristics of immune cells in pulmonary aspergillosis through single-cell sequencing, microbiome analysis, metabolomics, spatial transcriptomics, and other multi-omics approaches. We will analyze the role and regulatory mechanisms of immune cells in the pathological process of the disease. Through these analyses, we hope to provide a clearer understanding of how immune cells contribute to the immune response in pulmonary aspergillosis, offering theoretical and data support for the early diagnosis, precise treatment, and development of immune regulation strategies for the disease.

纳入标准:

1.肺毛霉菌病组:胸部影像学或气管镜检查提示存在肺部和(或)气管支气管感染,且经组织病理或宏基因组测序或镜检涂片提示毛霉菌感染; 2.单纯肺小结节组:胸部影像学提示肺部小结节(直径小于1cm)。

Inclusion criteria

1.Pulmonary Aspergillosis Group: Chest imaging or bronchoscopy suggests the presence of pulmonary and/or tracheobronchial infection, with histopathology, metagenomic sequencing, or microscopy smear indicating 2.Aspergillus infection Simple Pulmonary Nodule Group: Chest imaging indicates the presence of pulmonary nodules (less than 1 cm in diameter).

排除标准:

支气管肺毛霉菌病组: 1.具有支气管镜检查禁忌者; 2.拒绝气管镜检查者; 3.拒绝参加本项研究; 单纯肺小结节组: 4.具有支气管镜检查禁忌者; 5.拒绝气管镜检查者; 6.拒绝参加本项研究; 7.诊断为肺恶性肿瘤、肺间质纤维化、慢阻肺、肺栓塞及支气管扩张伴感染等呼吸系统疾病; 8.近1月内存在肺部感染及其它系统的感染。

Exclusion criteria:

Bronchopulmonary Aspergillosis Group: 1.Those with contraindications for bronchoscopy; 2.Those who refuse bronchoscopy; 3.Those who refuse to participate in this study. Simple Pulmonary Nodule Group: 1.Those with contraindications for bronchoscopy; 2.Those who refuse bronchoscopy; 3.Those who refuse to participate in this study; 4.Those diagnosed with lung malignancy, pulmonary interstitial fibrosis, COPD, pulmonary embolism, bronchiectasis with infection, or other respiratory diseases; 5.Those who have had a pulmonary infection or other systemic infections within the past month.
