研究目的：

主要目标： 1. 探究亮氨酸缺乏饮食对健康青年人群饮食偏好的急性调控作用和机理。 次要目标： 1. 探索亮氨酸缺乏饮食对压力、抑郁和焦虑倾向和相关行为的急性调控作用； 2. 探索亮氨酸缺乏饮食对人体代谢的急性调控作用和信号通路； 3. 探索亮氨酸缺乏饮食对人体微生物的急性重塑作用。  

Objectives of Study：

Primary Objective: 1. To explore the acute regulatory effect and mechanism of leucine deficiency diet on dietary preferences in healthy young people. Secondary objectives: 1. To explore the acute regulatory effects of leucine deficiency diet on stress, depression, and anxiety tendencies and related behaviors; 2. To explore the acute regulatory effect and signaling pathway of leucine deficiency diet on human metabolism; 3. Explore the acute remodeling effects of leucine deficiency diets on human microbes.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1. 身体质量指数（BMI）在18.5-24之间； 2. 年龄20-40周岁； 3. 总体健康状况正常，能够理解并遵守研究要求； 4. 自愿参与并签署知情同意书。

Inclusion criteria

1. Body mass index (BMI) between 18.5 and 24 kg/m2; 2. Aged 20 to 40 years; 3. In good general health with the ability to understand and comply with study requirements; 4. Voluntary participation with signed informed consent.

排除标准：

1. 有明确精神类疾病诊断史； 2. 过去两周内曾使用抗生素类、激素类药物；过去3个月内曾使用镇静催眠药物或其他抗抑郁抗焦虑、抗精神病类药物；过去3个月内酗酒和其他精神活性药物滥用史； 3. 嗅觉、味觉异常或过去1周内患急性感染性疾病、流感等短期影响嗅觉、味觉的疾病； 4. 有严重肝脏、肾、内分泌、心血管、胃肠道、肺、血液、代谢性疾病、癌症等疾病病史； 5. 有胃切除术、胃肠缝合手术、肠切除等消化系统大手术史（不包括阑尾炎）； 6. 现患传染病，如乙肝大三阳、活动性肺结核、艾滋病等； 7. 孕妇和哺乳期妇女； 8. 对干预所用奶粉成分过敏； 9. 血清亮氨酸水平低于80μM。

Exclusion criteria：

1. History of diagnosed mental disorders; 2. Use of antibiotics or hormonal medications within the past two weeks; use of sedatives, hypnotics, antidepressants, anxiolytics, or antipsychotics within the past three months; history of alcohol abuse or other psychoactive substance abuse within the past three months; 3. Abnormal sense of smell or taste, or acute infectious diseases (e.g., influenza) that may temporarily affect smell or taste within the past week; 4. History of severe liver, kidney, endocrine, cardiovascular, gastrointestinal, pulmonary, hematological, metabolic diseases, or cancer; 5. History of major digestive system surgeries such as gastrectomy, gastrointestinal anastomosis, or intestinal resection (excluding appendectomy); 6. Current infectious diseases, including hepatitis B "Da San Yang" (HBeAg-positive), active tuberculosis, or HIV/AIDS; 7. Pregnant or lactating women; 8. Allergy to any component of the intervention formula (milk powder); 9. Serum leucine level below 80 μM.

Intervention：

During the intervention period, subjects will receive leucine-deficient milk powder beverage (650-1000ml, to be consumed within 1 hour in divided doses) at breakfast (8:00-10:00), lunch (11:30-13:00), and dinner (17:00-19:00). Each meal will be accompanied by one apple. No other food is allowed. The amount of milk powder beverage is not restricted and will be provided as needed. Fasting is required after 20:00.

Eligible volunteers are randomized into intervention and control groups. Each volunteer is assigned a specific number, starting from 001. The numbers are arranged in no particular order and are generated randomly by computer. The randomization process involves the following steps: 1) Generating a random number for each participant included in the trial; 2) Sorting these numbers in ascending (or descending) order; 3) According to the pre-determined sample sizes of each treatment group, selecting the corresponding number of samples in the order of their random numbers and assigning them to different treatment groups.
